Effect Of Compound Acanthopanax Senticosus Powder On Immunity And Antioxidation In Mice

Abstract/Summary:
The aim of this study was to investigate the effect of Compound Acanthopanax Senticosus Powder on the immune and antioxidant function of mice diet.2 In the experiment,200 6-week-old unborn Kunming(KM)mice with close body weight and good health were selected.Preventive feeding for a week,after preventive feeding,the mice were randomly divided into 4 groups,namely control group,low-dose(0.20% Compound Acanthopanax Senticosus Powder)group,medium-dose(0.35% Compound Acanthopanax Senticosus Powder)group,and high-dose(0.50% Compound Acanthopanax Senticosus Powder)group.There are 5 repetitions in each group,and 10 repetitions per group.Feeding was carried out according to the added treatment for 1 to 21 days of the experiment,from the 22 nd day,all the basal diets were fed.The experiment period was 28 days.Two mice were randomly selected for each repeat on the 7,14,21,and 28 days They were sacrificed for sampling.On the 21 st day,another 2 mice were drawn from each replicate,injected with LPS(10 mg/kg),and sacrificed for sampling 8 hours later.The results showed as followed:1)Compared with the control group,the different treatments in each stage had no significant effect on the initial weight,the end weight,the average daily weight gain,the average daily intake and feed conversion rate(P > 0.05)2)On the 7th and 14 th days,the thymus index of the experimental group increased significantly(P < 0.05).Injection of LPS significantly increased the liver,spleen and kidney indexes of mice(P < 0.05),and prophylactic administration of Compound Acanthopanax Senticosus Powder could alleviate the increase in liver,spleen,and kidney indexes of mice caused by LPS(P > 0.05);There was a significant interaction between LPS and Compound Acanthopanax Senticosus Powder on liver index and kidney index of mice(P < 0.05).3)On the 7th day of the experiment,compared with the control group,the blood white blood count,lymphocyte number,median cell number,median cell ratio and hemoglobin content in the blood of the mice in Compound Acanthopanax Senticosus Powder group were significantly increased.On the 21 st day,the blood red blood count and platelet count of the low-dose group increased significantly,and the hemoglobin in the middle dose group was significantly increased(P < 0.05).On the 28 th day,the ratio of the median cells in the blood of the low-dose and medium dose groups was significantly increased,and the blood cell specific volume in the low dose group decreased significantly(P < 0.05).The platelet count of mice was significantly reduced by LPS injection(P < 0.05),and the preventive addition of Compound Acanthopanax Senticosus Powder could significantly increase the leukocyte count and significantly reduce the specific volume of blood cells in mice(P < 0.05).4)On the 7th and 21 st days,the IFN-γ content in the serum of the experimental group was significantly increased(P < 0.05);on the 14 th and 21 st days,the IFN-β content in the serum of the high-dose group was significantly increased(P < 0.05);on the 21 st and 28 th days,the IFN-α content in the serum of the experimental group decreased significantly(P <0.05).On the 14 th and 28 th days,the IL-1 content in the serum of the low-dose group decreased significantly(P < 0.05);on the 28 th day,the IL-2 content in the serum of the low-dose group decreased significantly(P < 0.05);on the 7th,14 th,21st and 28 th days,the IL-6 content in the serum of the experimental group decreased significantly,the serum IL-4content of the experimental group was significantly increased on the 7th and 14 th days(P <0.05);compared with the blank group,the TNF-α content in the serum of the experimental group decreased significantly on the 14 th,21st and 28 th days(P < 0.05).The contents of IFN-γ,IFN-β,IL-1,IL-8 and TNF-α were significantly increased by intraperitoneal injection of LPS,and the levels of IL-l0 were significantly reduced(P < 0.05).5)On the 14 th and 21 st days,the serum lysozyme and duodenal SIg A content of the test group mice increased significantly(P < 0.05).On the 21 st day,the content of β-defensin 2in the serum of mice in the test group was significantly increased(P < 0.05);the content of lysozyme in the serum of mice was significantly reduced by LPS injection(P < 0.05).Compared with the control group,prophylactic administration significantly increased the serum lysozyme,β defensin-2 and duodenal SIg A content in mice(P < 0.05);LPS and compound Acanthopanax Senticosus had a significant interaction effect on serum lysozyme,β-defensin-2,and duodenal SIg A content in mice(P < 0.05).6)On the 7th day,the serum T-AOC content of mice in each dose group increased significantly(P < 0.05),and the serum CAT content of mice in the middle and high-dose group increased significantly(P < 0.05).On the 14 th day,the low and high-dose groups were small The content of T-AOC in the serum of mice increased significantly(P <0.05).On the21 st day,the content of T-AOC in the serum of mice in the middle and high-dose group increased significantly,and the content of GSH-Px in the serum of mice in the low-dose group increased significantly.The MDA content in the serum of mice in the group reduced significantly(P < 0.05).On the 28 th day,the serum T-AOC content in the low-dose group increased significantly,and the serum CAT content in the low-and high-dose group increased significantly(P <0.05).MDA content in serum of mice injected with LPS increased significantly,and T-SOD content decreased significantly(P < 0.05);compared with the control group,the T-AOC content in the mice serum was significantly increased in the Compound Acanthopanax Senticosus Powder powder group(P < 0.05);the levels of T-AOC,T-SOD,CAT and MDA in serum of mice were significantly correlated with that of the LPS and Compound Acanthopanax Senticosus Powder(P < 0.05).In conclusion,it can be seen that the Compound Acanthopanax Senticosus Powder can enhance the body’s immunity and antioxidant function by regulating part of the body’s serum cytokines and antioxidant indicators,and has a certain continuation effect after all fed the basic diet.In addition,preventive administration of compound acanthopanax can alleviate the body damage caused by LPS.Under the conditions of this experiment,the appropriate level of Compound Acanthopanax Senticosus Powder in the diet of mice was 0.2% ~ 0.35%.
