Because of serious health hazards caused by Aflatoxin B1 contamination to humans and animals,it is important to evaluate the degrade degree of the mycotoxin in food safety and animal production.Therefore,this experiment aims to develop a green and non-polluting herbal compound as a feed additive against feed mold and prevent mycotoxin AFB1 from causing harm to the organism.The experiment was divided into two parts.(i)In vitro primary screening of Chinese herbal formulations against A.flavus,the evaluation of the inhibition effect of Aspergillus flavus was carried out on single aqueous extracts of Hibiscus Corticis Sytriaci,Fructus Cnidii,Bitter Ginseng,Cortex dictamni,Tangerine peel and Fructus kwhiae,respectively,and the ideal Chinese herbal medicine for the inhibition of A.flavus was then combined with SDA(sodium diacetate)and GOD(glucose oxidase)to screen the best inhibition compound according to the inhibition effect further.(ii)In vivo,Chinese herbal compound against aflatoxin AFB1 was taken,61-week-age laying hens were selected,and high and low concentration moldy feed groups were set up,and different kinds of compound preparations were added to the high and low dosage of moldy feed,including SDA&GOD group and Hibiscus Corticis Sytriaci&Bitter Ginseng&SDA&GOD group,and no addition was also used as the blank control group.The production performance,serum biochemical indexes,antioxidant performance,and serum antibody titers against avian influenza and Newcastle disease were evaluated after supplied moldy feeds to the normal diet for 28 d.The pathological histology was applied to evaluate the liver tissue damage in order to analyse the effect of different compound preparations,the data obtained were statistically analyzed to screen out the ideal combination of the drugs.Results:(1)In vitro,the results showed that the single Chinese medicine aqueous extract with 5 mg/m L of Hibiscus Corticis Sytriaci solution has the best obvious effect in inhibiting A.flavus,which was combined with SDA&GOD,and no growth of A.flavus colonies around the tablets was measured according to the paper method,and the inhibition rate was up to 100%;followed by SDA&GOD with 55.14 %,and the average growth diameter of A.flavus colonies was 8.14 mm.The AFB1 content in the 10% spiked group prepared according to ELISA assay was 58.9 μg/kg.(2)The results of the in vivo test showed that after adding the herbal compound against AFB1 toxin,the egg production rate of the herbal compound A3 group reached 91.27 %,which was significantly higher than the egg production rate of the A2 group with the addition of GOD+SDA at 15.08 %(P<0.05)and the A1 group without the drug was 34.52 %,the difference was obvious(P<0.05);In serum biochemical indexes,antioxidant properties and antibody titers of laying hens at different stages of the tested showed that with the increase of AFB1 toxin accumulation in laying hens,AST,ALP and ALT contents in each test group showed different degrees of decrease,while MDA contents in each group was increased,but SOD and GSH-PX in each group showed the significant decreasing trend.After vaccination of laying hens with Newcastle disease and avian influenza vaccines,the corresponding antibody titers in the high-dose AFB1 group decreased significantly,compared with the the blank control group(P<0.05);Pathological histological analysis showed that compared with the control group,the group with the addition of Chinese herbal medicine Hibiscus Corticis Sytriaci & Bitter Ginseng & SDA & GOD was significantly reduced in the damage to liver was caused by AFB1,compared with the other groups.In conclusion,the herbal compound preparation combined with the addition of Chinese herbal medicine Hibiscus Corticis Sytriaci & Bitter Ginseng & GOD & SDA in this experiment could effectively prevent and control the contamination of AFB1 in feed,and also has important clinical significance in promoting the healthy development of livestock and poultry.
