| In order to study the protective effect of Gynostemma pentaphyllum polysaccharides on oxidative stress damage in the small intestine of mice under heat stress,and to provide a theoretical basis for the further development and application of Gynostemma pentaphyllum polysaccharides.Mice aged 6 weeks were randomly divided into five groups: blank control group,HS group,GPP-L(50 mg/kg),GPP-M group(100 mg/kg),and GPP-H group(200 mg/kg).This study mainly used histological techniques and other research methods to observe and analyze the morphological structure of the duodenum,jejunum and ileum of groups of mice,as well as goblet cells,intraepithelial lymphocytes,argyrophilic cells and tight junction proteins ZO-1,OCLN and CLDN-1,and the total antioxidant capacity of each segment of the small intestine,the content of glutathione,malondialdehyde and lysozyme,and the activities of total superoxide dismutase,acid phosphatase and alkaline phosphatase were detected,and the results found that:1.Compared with the HS group,the mice in the GPP-L group gained weight at 20-21 d(P<0.05),and the mice in the GPP-M group increased their weight gain at 19-20 days(P<0.05);there was no significant difference in the small intestine length and small intestine index of the GPP group at different doses and the HS group.2.Different doses of GPP groups were able to alleviate morphological structure damage in the small intestine of mice under HS conditions.Compared with the HS group,villus height of segments of the small intestine of mice in the GPP-L group and jejunal of mice in the GPP-M group increased significantly(P<0.05);crypt depth of the GPP-L group and GPP-M group duodenum and jejunum in mice,and the GPP-H group mice with duodenum were significantly reduced(P<0.05),and the depth of the jejunal crypt in the GPP-L group and GPPM group was significantly higher than that in the HS group(P<0.05);the ratio of duodenal and jejunal V/C in the GPP-L group and GPP-M group increased significantly(P<0.05).3.Compared with the HS group,the number of small intestine segments in the GPP-L group and GPP-M group and the duodenal and jejunal cup cells in the GPP-H group increased significantly(P<0.05);the number of intraepithelial lymphocytes in the small intestine segments of the mice in the GPP-L and GPPM groups and the duodenal and jejunal epithelial lymphocytes in the GPP-H group was significantly reduced(P<0.05);the number of small intestine segments in the GPP-L group and the duodenal and ileal argyrophilic cells in the GPP-M group increased significantly(P<0.05).4.Different doses of GPP groups increased the expression of ZO-1,OCLN and CLDN-1 in various segments of the small intestine of mice under HS conditions.Compared with the HS group,the expression of tight junction protein ZO-1,OCLN and CLDN-1 in each segment of the small intestine in mice in the GPP-L group increased significantly(P<0.05);the expression of duodenal and ileal tight junction protein ZO-1 in mice in the GPP-M group increased significantly(P<0.05),the expression of jejunal and ileal tight junction protein OCLN(P<0.05),and the expression of CLDN-1 in each segment of the small intestine increased significantly(P<0.05);the expression of ileal connexin ZO-1in the GPP-H group increased significantly(P<0.05).5.Different doses of GPP groups significantly enhanced the antioxidant properties of various segments of the small intestine of mice under HS conditions.Between the GPP groups,the jejunal and ileal T-AOC activities of mice in the GPP-L group were significantly increased compared with the GPP-H group(P<0.05);the GSH content of each section of the small intestine in the GPP-L group and the GPP-M group was significantly higher than that in the GPP-H group(P<0.05);the ratio of duodenal and ileal GSSG/GSH in the GPP-L group and GPP-M group was significantly higher than that in the GPP-H group(P<0.05);the duodenal T-SOD activity of the mice in the GPP-L group and the GPP-M group was significantly higher than that in the GPP-H group(P<0.05);the MDA content of each section of the small intestine in the GPP-L group and the GPP-M group was significantly lower than that in the GPP-H group(P<0.05).6.Different doses of GPP groups can enhance the non-specific immunity of various segments of the small intestine of mice under HS conditions.Compared with the HS group,the duodenal and jejunal ACP activity of mice in the GPP-L group was significantly reduced(P<0.05),and the activity of jejunal and ileal AKP was significantly reduced(P<0.05);the duodenal and jejunal ACP activity of the mice in the GPP-M group was significantly reduced(P<0.05),the activity of the duodenal and ileal AKP was significantly reduced(P<0.05);The activities of ACP and AKP in the small intestine of mice in the GPP-H group were significantly reduced(P<0.05);the contents of LZM in the small intestines of mice in the GPP-L group,GPP-M group and GPP-H group were significantly increased(P<0.05).In summary,each dose group of GPP has a certain protective effect on the oxidative stress of the small intestine of HS mice,which can alleviate the morphological structure damage of various segments of the small intestine of mice,enhance the antioxidant and non-specific immunity of the small intestine,and the effect of the GPP-L group is better than that of other GPP groups. |
