Identification Of Laetiporus And Research On Artificial Domestication And Cultivation

There are abundant edible and medicinal fungi resources in China,and it is of great significance to develop and utilize wild edible and medicinal fungi for enriching edible and medicinal fungi resources in China.More and more people pay attention to the role of Laetiporus in pharmacology and health care.Many countries have begun to cultivate and commercialize Laetiporus.Due to the increasing demand for Laetiporus,the demand for Laetiporus is in short supply.In this paper,four Laetiporus strains were used as test materials,and identified by ITS molecular identification method,and the active components of four strains were determined,and then domesticated strains were determined;Single factor test and orthogonal test were used to optimize the culture medium and culture conditions of Laetiporus,and the original and cultivated species were screened,which laid a theoretical foundation and technical support for the development and utilization of this wild resource.The specific test results are as follows:1.Four Laetiporus strains were identified by ITS,and the results showed that LHJ,LH,Li were Laetiporus cremeporus,and Dai was Laetiporus sulphureus,which was consistent with the results of morphological identification;2.The content of polysaccharides,flavonoids,polyphenols and triterpenoids of Laetiporus(LHJ)was higher than that of the other three strains,and finally LHJ was the best domesticated strain among the four strains.3.The culture medium of Laetiporus(LHJ)mycelium was optimized by single factor and orthogonal method The optimum culture medium and conditions for Laetiporus(LHJ),were as follows:glucose 15g·L-1,yeast extract 2.0g·L-1,sodium chloride 0.7g·L-1,optimum temperature 30℃,optimum pH 7;4.Laetiporus(LHJ)grew fastest in formula 2(cottonseed hull 78%,wheat bran 20%,gypsum 1%,white sugar 1%),the growth rate was 0.517 cm·d-1,the full bottle time was 18 days,the hypHae were thick,white and dense,which was the best formula of the original culture medium for Laetiporus(LHJ);5.Laetiporus(LHJ)grew fastest with the formula of sawdust 43%,cottonseed hull 43%,wheat bran 10%,com flour 2%,gypsum 1%and white sugar 1%,the growth rate was 0.577 cm·d-1,the full bag time was 26 days,the hyphae were thick and dense,and the growth rate was significantly higher than that of other cultivars,which was the best formula of Laetiporus(LHJ).The prim ordia appeared in Formula 5,Formula 6,Formula 7 and Formula 8 after the hyphae were full of bags,which proved that domestication was successful.
