In this study,a strain of Pleurotus pulmonarius collected from Anji,Zhejiang Province was isolated and purified,and identified as Pleurotus pulmonarius by morphology and molecular biology.It belongs to Basidiomvcota,Agaricales,Pleurotaceae,Pleurotus.At the same time,this study further used the domesticated Pleurotus pulmonarius(S172)as the test strain,and Pleurotus pulmonarius No.2 as the control group to carry out the fruiting test under various media to study its physiological characteristics.The main results are as follows :(1)The results showed that sucrose and yeast extract as carbon source and nitrogen source,pH = 8.0,temperature 25 ℃ were suitable for the mycelial growth of S172.At the same time,the polysaccharide and protein contents of S172 and Xiuzhengu No.2 were compared.It was found that S172 had strong polysaccharide secretion ability and protein production ability under the same culture conditions.It shows that the wild strain has great medicinal potential.(2)The results showed that wild Pleurotus pulmonarius S172 had a complete extracellular enzyme system.Carboxymethyl cellulase,hemicellulase,amylase,laccase and other extracellular enzymes showed a certain activity during the cultivation process.The results showed that the mycelium growth rate of wild Pleurotus pulmonarius S172 was faster and the biological efficiency was higher under the substrate of sawdust and corncob.(3)In the cultivation of liquid mycelium,the growth of wild Pleurotus pulmonarius is stronger than Pleurotus pulmonarius No.2;the overall activity of hemicellulase and amylase was significantly higher;there was little difference in carboxymethyl cellulase activity between the two;the activity of laccase was stronger than that of Xiuzhen mushroom No.2 on the ninth day.Through the extracellular enzyme activity test of wild Pleurotus pulmonarius S172,we have a deep understanding of the enzyme system contained in it,including the function,metabolism and nutrition mechanism of the above enzymes,so as to provide a reliable basis for further study of its metabolism and nutrition mechanism,and this method has a wide range of applications and can be studied in many aspects.(4)In this study,S172 fruiting test was carried out under different substrates.The results showed that compared with rice straw and cottonseed hull,the mycelium growth rate of S172 was faster and the biological efficiency was higher under the substrate of sawdust and corncob.
