Platycodon grandiflorum is a perennial herb of Platycodon genus of Platycodonaceae family and has distinctive characteristics of Korean.It not only has outstanding medicinal and edible values,but also has strong ornamental value because of its upright and beautiful plant types,brilliant color and strong resistance,which is deeply loved and concerned by people.However,its color of flower is monotonous and mainly purple and white.The discovery of‘Fenhua’P.grandiflorum not only enriched the flower color,but also provided new germplasm resources for medicinal and edible use.However,‘Fenhua’P.grandiflorum often occurs color separation and can not maintain its distinctive color.The purpose of this study is to establish a rapid in vitro propagation system of‘Fenhua’P.grandiflorum by using stem tissue culture technology to preserve and propagate its germplasm,and provide basic materials for the application of‘Fenhua’P.grandiflorum in garden plants.The results are as follows:1.The optimal sucrose concentration for initiation culture of stems of‘Fenhua’P.grandiflorum was 40 g/L,the survival rate was 85.33%,and the fresh weight and dry weight were 5.54 g and 1.09 g,respectively.2.Axillary bud induction medium of‘Fenhua’P.grandiflorum was 1/2 N6+1 mg/L6-BA+0.5 mg/L IBA,the induction rate was 87.33%.3.The medium combination for axillary bud proliferation of‘Fenhua’P.grandiflorum was 3/4 B5+2 mg/L 6-BA+0.05 mg/L NAA,and the total nitrogen concentration in the medium was unchanged.The proliferation effect was the best,and the proliferation coefficient was 3.81.4.The rooting medium combination of‘Fenhua’P.grandiflorum was 1/2 B5+2mg/L IBA+0.5 mg/L NAA,and the rooting was the best when the total nitrogen concentration was adjusted to 3.35 mmol/L.The rooting rate was 70.67%,and the average number of roots per plant was 13.93.5.The substrate ratio of tissue culture seedlings of‘Fenhua’P.grandiflorum was perlite:vermiculite=1:2,and the survival rate was 81.11%.
