With the changes of modern life,People’s appreciation of phalaenopsis gradually favors miniature,Small flower,Double stem varieties.And gradually become the mainstream of Phalaenopsis market.In this experiment,3 varieties of micro double-stemmed Phalaenopsis were used as test materials.The test tube seedlings in different hormone concentration ratios,Under different activated carbon concentration treatments and different red and blue light quality ratios,Growth and proliferation of test-tube seedlings,And compared the responses of the cultivars to each treatment,So as to optimize the high-efficiency multiplication system of test-tube seedlings of various varieties.At the same time,Researches were carried out on the cultivation conditions required for the growth of miniature Phalaenopsis.Finally,For low temperature induction,The physiological responses exhibited by each variety under low temperature conditions were compared.Determining the nutritional requirements of the raw twin stems.The research results are as follows:1.Optimization of the efficient proliferation system of miniature Phalaenopsis test-tube seedlingsThe best medium for the three varieties are:’sweet too’ medium:MS+5.0 mg/L 6-BA+0.5 mg/L NAA+1.5 g/L activated carbon,’Pink Beauty’ medium:MS+7.5 mg/L 6-BA+0.5 mg/L NAA+2.0 g/L activated carbon,’Green Bear’ medium:MS+ 5.0 mg/L 6-BA+1.0 mg/L NAA+2.0 g/L activated carbon.The optimal ratio of red and blue light quality for sub-generation proliferation of test-tube seedling’sweet too’ is as follows,3:7,100%blue light,5:5,’Pink Beauty’:100%red light,7:3,’ Green Bear’:3:7.When the optimum activated carbon concentration of the test-tube seedling ’sweet too’ was 0.5 g/L,It was favorable for morphogenesis,When it was 1.0 g/L,It was favorable for root growth.At 1.5 g/L,The cultivar ’Pink Beauty’ was favorable for morphogenesis,And at 0.5 g/L,It was favorable for root growth.At 1.5 g/L of the ’Green Bear’ variety,It was favorable for morphogenesis and root growth.Test-tube seedlings are hardened and transplanted at a temperature of 25~30℃,light:12000 Lux,and water temperature:18~22℃.Survival rate as high as 99%.2.The influence of facility environmental factors on the flowering of miniature PhalaenopsisDuring the stem-throwing period,The ’Green Bear’ variety had good growth,Dwarf plant type,And fast pedicel growth.At the budding stage,The cultivar ’sweet too’ has plump plant shape,Many flower buds and early flowering.At the early flowering stage,The cultivar ’Pink Beauty’ is short in plant shape,Long in pedicel,With a large number of flowers and large corolla diameter,And is an outstanding variety in the early flowering stage.At the full blooming stage,the plant types of ’Pink Beauty’ and ’sweet too’ were dwarf and plump,With a large number of flowers and a small corolla diameter.Which were in line with the characteristics of miniature double-stalked Phalaenopsis.The concentrations of exogenous hormones that can be extracted from the three varieties are as follows:’sweet too’ was the best in 6-BA 150 mg/L,Followed by GA3 150 mg/L and KT-30 150 mg/L.’Pink Beauty’ was the best in 6-BA 150 mg/L,Followed by KT-30 150 mg/L.’Green Bear’ was optimal at 6-BA 200 mg/L.The substrates suitable for the growth of the three varieties are sphagnum moss and coconut bran,And peat is not conducive to growth.3.Physiological changes of microphalaenopsis before and after double stem and flower bud differentiationThe varieties with stronger chlorophyll,Proline and antioxidant enzyme activities in the process of stem-raising at low temperature were ’Green Bear’,’Pink Beauty’,and ’sweet too’.When flower bud differentiation occurs,The order of soluble protein content is:’Pink Beauty’,’Green Bear’,And ’sweet too’.To sum up,By changing the hormone concentration,Light-quality ratio,Etc.the test-tube seedlings can proliferate efficiently.The environmental factors of the facility can improve the stem-emergence rate.
