Established The Reference Standard Of Bama Miniature Pigs’ Blood Physiological And Biochemical Indicators And Screened Abnormal Individual Of Blood Parameters In Enu-induced Bama Miniature Pig Mutants

Bama miniature pigs have irreplaceable advantages than other animals in the place of genetic,development, disease models and organ xenotransplantation, and are widely used in the study of thecardiovascular system and heterogeneous organ transplantation. The hematologic and biochemicalof Bama miniature pigs are important basic indicators in pathology, physiology and toxicologyresearch work, and are also an important basis to determine the physiological state of Bamaminiature pigs. But the feeding standards of Bama miniature pigs have not united because they arean emerging large-scale experimental animal, many research departments take restricted feeding onBama miniature pigs to prevent fleshing, which may interfere the determination of physiologicaland biochemical indicators. Genetically modified by Chemical mutagen ENU is a new method toprepare animal models of disease. It is slso faster, higher intensity of mining gene and produced ahuge number of mutant animals a short time than traditional gene knockout and trangenosis. Due tothe randomness and uncertainty of ENU mutagenesis, techniques and methods of screening mutantanimal models of disease are particularly important. It is not only well improve and supplement thescreening methods of animal models of disease, but also can obtain individual of abnormal bloodparameters through the screening on the blood physiological and biochemical indexes of F1generation of Bama miniature pigs. In our study,336weaning Bama miniature pigs were randomlydivided into control group,80%of the diet group and50%of the diet group according to thedifferent restricted feeding levels, blood samples were collected from precava at60daysage and90daysage, seven important hematologic parameters and ten physiological parameters weredetermined, then we counted the differences of indicators under different restricted feeding levels,we got the underlying data of Bama miniature pigsblood physiological and biochemical indexes,and establish a reference range of physiological and biochemical indicators. Then seven F0generation boars were obtained by ENU mutagenesis, after mating with wild-type sows, we got 1101F1theoretically carry genetic mutant gene individuals. And then through the determination ofits physiological and biochemical index, repeat testing the individuals on blood index value exceedsstatistics Mean±2SD, we got five individual of abnormal blood parameters. Results are as follows:1. Establish the reference standard of Bama miniature pigsblood physiological andbiochemical indicators.In our study, there was a positive correlation with the restricted feeding levels and theindicators such as ALT, TP, UREA, CREA, RBC, HGB, HCT, MCH and MCHC. There was also anegative correlation with the restricted feeding levels and the indicators such as TCHOL, GLU,PLT, MCV, RDW-SD and RDW-CV. The blood physiological and hematologic parameters ofBama miniature pigs have a significantly different in three restricted feeding levels. At the sametime, due to the omnivorous of Bama miniature pigs, appropriate green fodder is consonant with itsgrowth and physical needs, and has a positive effect on establishment of Bama miniature pigsblood biochemical standard parameters. We can get a stable and standard blood physiological andhematologic parameters under20%restricted feeding in this study, and the indexes under20%restricted feeding can become the reference range of Bama miniature pigsblood physiological andbiochemical indicators.2. Abnormal individuals of blood physiological and biochemical indexes in F1generation.We used the F0generation boars to mating with wild-type sows, and we got the F1genetationindividuals, then strictly screening of blood physiological and biochemical indexes in the F1generation individuals, recheck the individuals whose index value more than statistics Mean±2SD,finally we found five abnormal individuals. In those abnormal individuals, No.006002()individuals MCV(71.5fL72.3fL;70.8fL), No.027098() individuals PLT(779×109/L791×109/L;783109/L), No.006009() individuals ALT(116u/L;131u/L;124u/L) No.006111()individuals TG(1.89mmol/L1.64mmol/L;1.21mmol/L)and UREA(12.3mmol/L13.8mmol/L;10.0mmol/L)are more than statistics Mean+2SD, whats more, they are also higher than normalBama miniature pigsphysiological and biochemical index reference value. But the No.027020()individuals TP(38g/L46g/L;40g/L)is less than statistics Mean-2SD, and also lower than the TPin normal Bama miniature pigs. So we preliminary judgment they may be dominant mutationindividual.In summary, this study we discussed the influence of restricted feeding levels on physiological and hematologic parameters, and established a reference range of Bama miniature pigsbloodphysiological and biochemical indicators. Then we measured and screened the blood physiologicaland biochemical parameters strictly of F1generation individuals who had been mutation by ENU,we found five abnormal individuals. Our study provided genetic material for the furtheridentification of dominant mutation individuals as well as establishment of disease pedigrees underENU-induced.
