Main Active Constituents And Activity Evaluation Of Paeonia Ostii Fruit Around Its Full Development Stage

Abstract/Summary:
As an important oil crop,peony has attracted much attention for its fatty acid content.However,some wastes produced in the production of peony seed oil,such as fruit pod,seed shell and seed cake,have not been fully utilized,so it is particularly important to study the natural phytochemical composition of peony fruit.This study firstly P.ostii fruit phenotypic traits of different periods were determined,the P.ostii fruit pod and seed a grease part compound changing law and its biological activity,through the mass spectrometry(UPLCQ-TOF-MS/LC-ESI-:QQQ-MS)combined with molecular network(MN),revealing the metabolites of P.ostii fruit completely,and using the multivariate analysis methods to screen potential differences metabolites,finally,stilbene compounds in the seed shell of P.ostii response surface process was optimizated,the results are as follows:1.The diameters of pod and seed increased at the early stage of fruit development and then tended to be stable with fruit ripening.During the ripening process,the pericarp changed from crab yellow to dehiscence,and the seed coat turned dark brown to black.The water content of seeds has been decreasing since the beginning,and then decreases sharply after S8(8,July),that is,the water content of seeds decreases rapidly during the rapid maturity period.The changes of oil content and water content of seeds are diametrically opposite.2.The content of total phenols in pods was significantly higher than that in seeds.During the development of tree peony fruits,the total phenol content in fruit pods and seeds showed a completely opposite trends,the former decreased and the latter increased.In addition,the contents of total flavonoids and flavanols in seeds were significantly lower than that in pods.By UPLC-Q-TOF-MS combination of GNPS screening identified 42 kinds of metabolites,mainly polyphenols,flavonoids,monoterpene glycosides styrene and 2 class is given priority to,quantitative LC/ESI-:QQQ-MS of 34 compounds,different development phases have different compounds,among them with the highest content of paeoniflorin class,at the same time,resveratrol mainly exists in the peony seed,up to 1524.66 mg/kg;The results of PCA analysis showed that there was an obvious separation trend in the distribution of seed and pod compounds.Combined with the prediction of VIP by OPLS-DA analysis,18 potential characteristic compounds were screened out.3.Fruit pods and seeds have certain antioxidant activity and antimicrobial activity,and there were significant differences,pod clearing DPPH free radical ability decline after rising first,contrary to seed change rule,ABTS radical ability change tendency is alexandrine,best removal ability and pod were appeared in the period of S1(28,April),both FRAP reducing power change trend is consistent,after rising to decline and in S4(28,May)period to achieve the best;Correlation analysis showed that there was a significant positive correlation between total phenol content and antioxidant capacity in fruit pod and seed extracts.The fruit pods and seeds had better bacteriostatic effect on Proteus vulgaris and Listeria monocytogenes than on Escherichia coli and Staphylococcus aureus.4.Stilbene contains abundant stilbene in peony seed shell.Single factor results showed that Suffruficosol B and trans-gnetin H contents in seed shell were significantly higher than Resveratrol,and trans-gnetin H content was the highest.The optimal extraction process of Suffruficosol B was obtained by response surface optimization as follows: ultrasonic power60 W,temperature 50 ℃,ultrasonic time 40 min,solid-liquid ratio 1:15,ethanol concentration70%.The optimal extraction conditions of Trans-Gnetin H were as follows: temperature 60 ℃,ethanol concentration 90%,solid-liquid ratio 1:40,power 80 W,time 50 min.
