Paeonia ostii ‘Fengdan’ is the main tree peony cultivar in the oil cultivation system. Its seed production and seed oil content are higher. The quality of seed oil is better.However, Paeonia ostii ‘Fengdan’is a cross pollinated plant and seed setting characteristics were poor, it is necessary to configure appropriate pollination cultivar in order to improve the yield of Paeonia ostii’Fengdan’. Paeonia ostii ‘Fengdan’ and 21 kinds of Paeonia suffruticosa Zhongyuan Group were choosed as test material, by observing and recording Flowering phenology and the secretion of stigma mucus and determining pollen viability and stigma, and then select the cultivars of high pollen viability(pollen germination rate >50%) to carry out artificial pollination experiment and Using the pollen of Paeonia ostii’Fengdan’ to carry out the artificial pollination experiment on different cultivars of Paeonia suffruticosa Zhongyuan Group.The percentage of fruiting, seed number per frui and single seed weight were counted and the main results are as following:1. By observing and recording Flowering phenology, ‘Cao Zhou Hong’, ‘Bing Ling Zi’, ‘Hong Bao Shi’ and ‘Feng Dan’flowering completely meet, is suitable in flowering pollinationcultivars; ‘Hong Xia Ying Yue’, ‘Xue Ying Tao Hua’, ‘Wu Long Peng Sheng’, ‘Zi Er Qiao’, ‘Yin Hong Qiao Dui’, ‘Chun Hong Jiao Yan’, ‘Mo Chi Jin Hui’, ‘Yan Hong Jin Bo’, ‘Fen Lan Pan’, ‘Cong Zhong Xiao’, ‘Ya Pian Zi’, ‘Zi Ju’, ‘Da Hong Bao Zhu’, ‘Zi Die Fei Wu’, ‘Man Yuan Chun Guang’, ‘Fei Cui He Hua’ and ‘Ge Jin Zi’for both artificial pollination was suitable for mining powder, the pollination tree, natural pollination;’Bai Xue Ta’flowering stage and Paeonia ostii‘Fengdan’flowering stage of’ nonoverlapping period, suitable for artificial pollination pollen collection.2. By the I2-KI staining method, TTC staining and in vitro germination method to determine pollen viability, the results show, TTC staining, I2-KI staining were unable to determine the pollen vitality, in vitro germination method is most suitable for the determination of pollen vitality test method. In vitro germination method, determination of pollen viability results shows that there is significant difference for 21 kinds of Paeonia suffruticosa Zhongyuan Group’s pollen vitality and pollen germination rate, the highest reaches 91.3%(‘Fen Lan Pan’) and the lowest is only 3.6%,(‘Chun Hong Jiao Yan’).3. By observing the stigma of mucus secretion and the stigma receptivity and stigma receptivity and mucus production in showed a trend of first increased and then decreased, stigma can be granted early start time to the mucus appear, termination time later than mucus disappeared, mucus secretion can as a flower column head can be granted a concise way of judging and artificial pollination can reference the stigma of mucus secretion.4. Paeonia ostii ‘Fengdan’and other herbaceous plants are the same, there is no apomictic reproductive phenomenon, dominated by cross pollination, weak self fruitfulness.5. Through pollination test statistics of the seed setting rate, average seed number and average grain weight, then calculate the average seed weight of single flower pollination and weight gained rate, taking Paeonia ostii ‘Fengdan’natural pollination as control, ‘Ya Pian Zi’, ‘Zi Ju’, ‘Fen Lan Pan’ and ‘Hong Bao Shi’’s average seed weight of single flower pollination and pollination varieties was significantly higher than that of’ natural pollination of Paeonia ostii ‘Fengdan’, the weight gained rate up to 26.7%~72.5%, they can be used as testing cultivar of Paeonia ostii ‘Fengdan’ to plant. The average seed number of pollinated flowers by single evaluation of different Paeonia suffruticosa Zhongyuan Group cultivar was granted to Paeonia ostii ‘Fengdan’’s pollen after seeding, results showed that the average seed number of single pollinated flowers added from the ‘Ya Pian Zi’ and ‘Mo Chi Jin Hui’ grain to 12.7 grains, 6 cultivars of 8 cultivars’(‘Cao Zhou Hong’, ‘Mo Chi Jin Hui’, ‘Ya Pian Zi’, ‘Man Yuan Chun Guang’, ‘Da Hong Bao Zhu’, ‘Cong Zhong Xiao’) fecundity were significantly improved. |
