| Ⅰ.Physiological Function of Colored RiceA large number of studies have shown that black rice has strong physiological functions,including antioxidant,lowering blood sugar and other physiological functions.However,there is no report on the comparison of physiological functions between different colors of rice,so this study conducted blood glucose and lipid experiments on different colors of rice to study the physiological functions of different colored rice cultivars.The results were as follows:1.The combination of streptozotocin and high-fat feed could cause type 2 diabetes in mice;the intervention of red rice materials in diabetic mice can reduce their blood sugar and blood lipids.2.Red rice and black rice could reduce the blood lipid level of mice with acute hyperlipidemia,but did not affect the weight of mice.Ⅱ.Functional Study of Gene Os05g0320700 involved in Flavonoid biosynthase pathway in RicePigmented rice is an important functional rice because of its rich flavonoid components.The study performed the transcriptome sequencing analysis of three different colors of rice in order to explore the improvement of quality.The gene Os05g0320700 was found to play an important role in increasing the content of flavonoids.The gene is a cytochrome P450,presumably encoding cinnamate 4-hydroxylase(C4H),and the gene was temporarily named OsC4H.C4H is the second key enzyme in the phenylalanine metabolism pathway,and its metabolite coumaric acid enters the lignin synthesis and flavonoid synthesis pathways.In order to explore the effect of this gene on the flavonoid biosynthesis pathway and the physiological function of red rice,the over-expressed plant OsC4H-OE and the knockout plant OsC4H-KO were constructed using red rice Hong Nuo as transgenic receptors.The results are as follows:1.The 1000-grain weight increased in overexpressing plant OsC4H-OE than that of wild type,and the leaf epidermis appears fluff;the knockout plant OsC4H-KO was shorter than the wild-type plant,the panicle length becomed shorter,and the 1000-grain weight did not change than wild type.2.The expression of OsC4H in panicles and stems was higher than that in leaves and roots;the expression of photosynthesis-related genes and flavonoid synthesis genes in OsC4H-OE young panicles was higher than that of Hong Nuo,and OsC4H-KO was lower than that of Hong Nuo.3.The content of polyphenols and flavonoids in the extract of colored rice peel were the highest in OsC4H-OE,followed by Hong Nuo,and the lowest in OsC4H-KO;the antioxidant capacity(ORAC)of the peel extract was similar to that of flavonoid,and the antioxidant capacity were ordered:OsC4H-OE> Hong Nuo > OsC4H-KO.4.OsC4H directly or indirectly regulated expression of flavonoids in the rice peel and affect the physiological function of rice. |
