| Soybean is one of the important crops.In the context of the urgent need to revitalize China’s soybean industry,improving the vitality of soybean seeds can reduce production costs and increase the economic benefits of farmers.In this study,the recombinant inbred lines,which was derived from Gong 387-1 and Ya’an Heidou,were used to test the phenotypes of seed vigor related traits(germination energy,germination percentage,germination index,vigor index,seedling height,mean germination time,conductivity,the time of the germination percentage reaches 50%,protein content,oil content and field germination percentage)of newly harvested seeds and artificially aged seeds in different planting years in 2018 and 2019.Based on the BSA analysis method and the whole genome resequencing technology,the different regions of seed vigor related traits were detected.Indel markers were designed for QTL location analysis of soybean seed vigor related traits.The main results were as follows:1.In the newly harvested seeds and the aged seeds,most of the seed vigor correlation differences of parents were significant or extremely significant.Among the newly harvested and aged seeds,RIL population has rich variation in seed vigor related traits,super parent separation is common,and the distribution of each trait is normal,which conforms to the genetic characteristics of quantitative traits.The correlation difference of seed vigor of RIL population was very significant among families.The correlation between new seeds seed vigor in RIL population,such as weight,conductivity,was significant or extremely significant in different years;the traits of weight,conductivity,protein content,oil content were significant or extremely significant in the interaction between years and families;the weight in RIL population was significant in the interaction between years,years and families in aged seeds.2.Based on the BSA analysis method,through the whole genome resequencing of four DNA samples(Gong 378-1,Ya’an Heidou and two extreme mixed pool),we got six different regions related to seed vigor on six chromosomes,and designed indel markers.According to the six different regions,38 pairs polymorphic indel markers were screened for QTL.3.A total of 20 QTLs related to seed vigor were detected by ICIM,7.93%~27.92% of the phenotypic variation was explained.In newly harvested seeds,17 QTLs related to seed vigor were detected,which accounted for 7.93%~27.92% of the phenotypic variation.Among them,q FGP-2-1,which controls the field germination percentage,and q W-2-1,which controls the weight,form QTL clusters on chromosome 2.q P-5-1,which controls the protein content,and q O-5-1,which controls the oil content,form QTL clusters on chromosome 5.In two years,q VI-6-1 was located to control vigor index,which was close to each other,forming a QTL cluster.In two years,q GI-2-1 was repeatedly located to control germination index.Three QTLs related to seed vigor were detected in aged seeds,which explained 12.77%~17.91%of phenotypic variation. |
