Inonotus obliquus as a precious medicinal fungi has a variety of medical and health care functions aroused people’s attention,and has been used to treat cancer,heart disease,fall blood sugar for a long time.Since the sixteenth century,Inonotus obliquus has been used as a folk remedy to prevent and cure various of diseases in East Europe,Russia,Poland and Finland,etc.Triterpenes are the main active ingredient in the Inonotus obliquus.Squalene synthase is a key enzyme in the synthesis of triterpenes,catalysing the conversion of FPP to squalene.Squalene as a precursor synthetizes triterpenes,sterols,cholesterol and other terpenes.The activity and content of Squalene synthase determines the production of triterpenes.Triterpenes have many functions in health protection and physiology,so much attention has been paid to the research of Squalene synthase.In this research,we use the Inonotus obliquus LNUF008 which is kept in the laboratory of Liaoning University as the activity strains.Firstly,design the degenerate primer based on the high homology squalene synthase of other species.Secondly,amplify the partial sequences of the squalene synthase of Inonotus obliquus with PCR.Thirdly,design the specific primers based on the known sequence of Inonotus obliquus,get the complete sequence of squalene synthase by a combination of RT-PCR and RACE.The location and cloning of the squalene synthase gene from Inonotus obliquus laid a fundation for the further study of the active ingredients in the Inonotus obliquus.With the value of Inonotus obliquus widely known by people,Inonotus obliquus resource has been constantly consumed.Currently the Inonotus obliquus can not be cultured,resulting in the fruiting bodies facing depletion.In view of the status of the fruiting body from Inonotus obliquus,researchers have begun cultivating the mycelia of Inonotus obliquus,Trying to find the alternatives of the fruiting body.Extracted triterpenes from the fruit bodies and mycelium of Inonotus obliquus,and determined their contents and effects of bacteriostasis.The experimental results showed that the feasibility and practicality that the mycelium replace the fruit bodies of Inonotus obliquus to product the active substances,which provides a new way for the development of the Inonotus obliquus.
