According to the biological classification,the Inonotus obliquus belongs to the family of Polyporaceae,brown Poria,and it is a kind of rare medicinal fungus.It was showed that,the main medicinal components of Inonotus obliquus was polysaccharide,in which had the value on the treatment for cancers,improving immunity, anti-aging,cardiovascular and cerebrovascular diseases. In the recent more than 10 years, the research on Inonotus obliquus is gradually extensive.Chaga has been one of the most valuable development in medicinal fungus.As a kind of fungus living in frigid zone,wild Inonotus obliquus is extremely rare,collecting wild Inonotus obliquus to extract active substances is impossible for large-scale production.The artificial domestication cultivation of Inonotus obliquus has not been realized at present,so extracting active substances from artificial cultivation of Inonotus obliquus is not available.Therefore,we may get mycelium through deep liquid fermentation culture of Inonotus obliquus,from which we extract active substances,and it is the best way for large-scale production of Inonotus obliquus at present.The research on deep liquid fermentation culture of Inonotus obliquus was made by pilot-scale fermentation tank with the volume of 100L.The best carbon and nitrogen source,the initial pH of fermentation liquid and the best process conditions of fermentation was studied,which provided the basis for large-scale fermentation production of Inonotus obliquus mycelium.The best conditions for deep liquid fermentation culture of Inonotus obliquus mycelium were determined by the single factor experiments and response surface experiments.First,through the single factor experiments between several common carbon sources,the production of Inonotus obliquus mycelium was contrasted,and the best carbon source for deep liquid fermentation culture of Inonotus obliquus mycelium was determined according to the production and the cost of materials.Then,through the single factor experiments about the concentration of carbon sources,the production of Inonotus obliquus mycelium between different carbon sources was contrasted,on which the best concentration of carbon source in the single factor conditions was found.The best concentration of nitrogen source under the best kind of nitrogen source and the single factor was determined by the single factor experiments about the type and the concentration of nitrogen sources in the same way.The single factor about initial pH was also made to determine the best initial pH in single factor conditions.According to the results of the single factor experiments,the orthogonal experiment was designed by response surface method,in which the concentration of carbon sources and nitrogen sources and the initial pH were used as three factors and three levels,and the best process conditions for deep liquid fermentation culture of Inonotus obliquus mycelium were determined.Through the experiments,the waste molasses with the concentration of 236.4mL/L,was the best carbon source and the powder of soybean cake with the concentration of 98.6g/L,was the best nitrogen source.The best initial pH was 6.1 and the temperature of fermentation was 26℃,which lasted for 12 days.The production of Inonotus obliquus mycelium could achieve the most 9.7g/L with the conditions listed above.
