| Inonotus obliquus(Fr.)?i\at has been known for more than one hundred years as a medicinal fungus in folk medicine in Russian, it can cure many difficult diseases. Since Humfeld American H and other workers studied submerged culture of mushroom and put forward to culture mycelia of mushroom by this method, fungi have been cultured successful. But the researches in Inonotus obliquus are scant in this aspect; furthermore, the extraction and separation researches of polysaccharides, which were obtained from submerged culture, are rare.In this paper, obtained Inonotus obliquus culture was used in fermentation. To get more MDW and polysaccharides, both the one-factor-at-a-time and the SAS methods were applied to obtain optimum media for them. The mycelia dry weight (MDW), the residual sugar, exo-polysaccharides (EPS) and Triterpenoid substances in the media were detected by Dry weight method, DNS method, Phenol - sulfuric acid method, Vanillina-Glacial acetic acid method. By the extraction separation of Inonotus obliquus polysaccharides, two rarefied polysaccharides can be obtained, IO2b, IO3a. The results were summarized as follows:(1)The optimum media of MDW: Sugar corn syrup 2.76%, oluble starch 1.9%, Yeast powder 0.75%, epetide 0.95%, H2PO4 0.2%, MgSO4 0.05%; The best accumulated time of MDW is 6d; The best culture temperature is 28℃; the best initial pH is 5; the best shake speed is 130r/min; seed quantum is 15%; liquid quantum is 70mL/250mL.(2)The optimum media of polysaccharides: Sugar corn syrup 2.1%,Soluble starch 2.23%, Yeast powder 0.51%, pepetide 0.41%, KH2PO4 0.2%, MgSO4 0.05%; The best accumulated time of MDW is 7d; The best culture temperature is 28℃; the best initial pH is 6; th best shake speed is 130r/min; seed quantum is 15%; liquid quantum is 70mL/250mL.(3)After fermenting of Inonotus obliquus, mycelium and fermentation liquor was detached. Fermentation liquor was concentrated, deposited in alcohol, removed protein, dialysis, and evaporated, then Inonotus obliquus exo-polysaccharides IO was obtained. After the stage separation of DEAE-52 and sephadex G-200, Inonotus obliquus exo-polysaccharides IO2b and IO3a was obtained.(4)The monosaccharide constituent of Inonotus obliquus exo-polysaccharides include glucose, galactose, mannose and Arabinose. Infrared Spectroscopy shows Polysaccharide absorption peak. By HPLC method, molecular weight of IO2b is 1.64×105, molecular weight IO3a is 8.26×103.
