Excessive energy intake and the imbalance of metabolic function are important factors leading to the occurrence and development of obesity,while long-term obesity state will become the "medium" of a variety of chronic diseases,causing the occurrence of a variety of chronic diseases in human body.The functional components of plant-derived tea,lotus leaf and hawthorn extracts have been reported to exert lipid reduction and weight loss through various pathways,including inhibiting adipocyte genesis and regulating mitochondrial metabolism.This study used tea functional ingredients(EGCG,theaflavin,Fu tea extract),lotus leaf extract and hawthorn extract to explore four different fat reduction formula(formula 1(G1): lotus leaf extract + hawthorn extract;formula 2(G2): EGCG 90% + lotus leaf extract + hawthorn extract;formula 3(G3): thea-flavin 50% + lotus leaf extract +hawthorn extract;formula 4(G4): Fu tea extract + lotus leaf extract + hawthorn extract)on obese mice induced by high fat diet(HFD).The main results are as follows:1.The four fat-lowering weight loss formulas can effectively prevent body weight gain in mice on an HFD diet and reduce the body weight growth rate.The weight growth rate of model group was more than 30%,22.6% in G1 group,20.3% in G2 group,20.7% in G3 group,21.7% in G4 group;and compared with model mice,liver,epididymal fat and perirenal fat weight and fat cell area in different formulas were down-regulated to varying degrees.2.Four fat-lowering formulas inhibited AST and ALT content in the liver,TG,TC,LDL-C and HDL-C content in serum,and the levels of inflammatory factors(IL-1,IL-6,I L-6 and INF-γ)in vivo were also significantly down-regulated;3.Transcriptomics suggested that the four formulations played a role in HFD-induced obesity by regulating liver function.Energy metabolism function of liver and redox pathway were mainly regulated in G1 group,oxidative phosphorylation of liver,thermogenic pathway,and adipogenesis pathway in G2 group were regulated,G3 group had regulatory effects on liver inflammation and immune pathways as well as MAPK pathway,liver insulin resistance,glyceride metabolism signaling pathway were regulated in G4 group;4.The results of intestinal microbiota sequencing showed that the abundance and composition of intestinal microbiota were affected by different lipids and weight loss formulas,which mainly regulated Akkermansia,Oscillospira and Clostridiales.
