Alcoholic liver disease is the most common chronic liver disease in the world,longterm drinking can cause imbalance of the body’s antioxidant system,abnormal accumulation of lipids in the liver,and a series of pathological reactions such as liver cell damage,necrosis,and liver cirrhosis.Studies have shown that tea polyphenols have multiple functions such as anti-inflammatory,antioxidant,anti-alcoholic liver fibrosis,antibacterial and intestinal health protection.Ampelopsis grossedentata and Cassia seeds have the functions of lowering blood pressure,regulating blood lipids,improving eyesight,moistening the intestines,protecting the liver and inhibiting bacteria.In this study,five extracts were selected to prepare solid beverages for cell test and animal test of alcoholic liver injury,transcriptomics and highthroughput sequencing of intestinal flora were used to analyze the regulatory effect and potential mechanism of tea solid beverage on chronic alcoholic liver injury in rats,it provides a scientific basis for the prevention and treatment of chronic alcoholic liver injury,the research and development of tea solid beverages and the utilization of tea extracts.The main results are as follows:(1)Tea solid beverage had significant regulatory effect on chronic alcoholic liver injury in rats.Formula 1: Tea Polyphenols + Ampelopsis grossedentata extract + Cassia seed extract(1:3:1)and formula 2: Ampelopsis grossedentata extract + Cassia seed extract(1:3)were screened out by cell test.According to the results of cell tests,animal tests showed that,the tea solid beverage group could effectively improve liver cell damage caused by alcohol,and effectively reduce the production of fat vacuoles and inflammatory cell infiltration in a dose-dependent manner.Tea solid beverage could effectively alleviate the increase of AST,ALT content,LDH enzyme activities and the expression of inflammatory factors TNF-α and IL-6,and significantly enhanced GSH,TG concentration and SOD activity and decreased MDA concentration in liver caused by drinking alcohol.(2)Transcriptomic analysis speculated the potential mechanism of tea solid beverage alleviating chronic alcoholic liver injury in rats.Transcriptional analysis of CON group,ETOH group and H1 group showed that tea solid beverage intervention in drinking rats mainly increased the synthesis and degradation of ketone body by up-regulating the differential gene Hmgcs1 and down-regulating the differential gene Pfkfb1 to reduce glucose metabolism and protect the liver.(3)High-throughput sequencing of intestinal flora in CON group,ETOH group and H1 group showed that tea solid beverage intervention in drinking alcohol rats mainly increased the abundance of Roseburia、 Coprococcus and norank_f__Butyricicoccaceae,and regulated pathways such as mineral absorption,bile secretion,adipocytokine signaling,and peroxisomes to balance the gut microbiota,which in turn had a protective effect on the liver.
