Numerous studies show that probiotics play many physiological effects,such as the intestinal flora regulation,immune improvement,and lipid-lowering function.Based on the preliminary work of our laboratory,this paper prepared and evaluated the in vitro performance of the Bifidobacterium breve beads and Lactobacillus plantarum-Lactobacillus fermentum-Bifidobacterium breve compound beads.The in vivo performance of these beads were also studied for their lipid-lowering effects using obese mouse.The following is the process and results for preparation and in vitro characterization of the Bifidobacterium breve beads and Lactobacillus plantarum-Lactobacillus fermentum-Bifidobacterium breve compound beads: The low methoxyl pectin(LMP)was used as wall material,Bifidobacterium breve,Lactobacillus fermentum and Lactobacillus plantarum alone or in combination were encapsulated into the beads by ionic-crosslinking process.The encapsulating rate was higher than 99%.The in vitro tests in simulated gastrointestinal fluid showed 4.82 and1.76 log(CFU/g)decrease for unencapsulated Bifidobacterium breve and the Bifidobacterium breve beads,respectively,and 4.82 and 1.74 log(CFU/g)decrease for unencapsulated Lactobacillus plantarum-Lactobacillus fermentum-Bifidobacterium breve and the Lactobacillus plantarum-Lactobacillus fermentum-Bifidobacterium breve beads,respectively.The storage stability test showed the protective effect of LMP beads on Bifidobacterium breve,Lactobacillus fermentum and Lactobacillus plantum.The procedure and results of study on the effect of probiotics beads on lipid-lowering in obese mice with high fat diet are detailed as follows: The mice were fed with normal diet and high fat diet,respectively for establishing high-fat mouse model.Then these mouse were divided into six groups: Blank control(Group A,fed with base chow),high-fat control(group B,fed with high-fat chow),high-fat positivecontrol(group C,fed with high-fat chow plus Oristat),Bifidobacterium breve beads-fed group(group D,fed with high-fat chow plus Bifidobacterium breve beads),Lactobacillus plantarum-Lactobacillus fermentum beads-fed group(group E,fed with high-fat chow plus Lactobacillus plantarum-Lactobacillus fermentum beads),Lactobacillus plantarum-Lactobacillus fermentum-Bifidobacterium breve beads-fed group(group F,fed with high-fat chow plus Lactobacillus plantarum-Lactobacillus fermentum-Bifidobacterium breve beads).The results showed that the probiotics beads reduced the TC and TG contents in obese mice serum.The intervention of different probiotics beads reduced the contents of IL-6,IL-2β,LPS and TNF-α in high-fat diet mice serum,alleviated the degree of liver cell steatosis,reduced the size of adipocytes,and protected the intestinal epithelial cells from pathological changes in obese mice.High-fat diet significantly changed the structure of mouse intestinal flora,the proportion of Firmicutes increased significantly,while the probiotics beads intervention adjusted the flora structure,increased the number of bifidobacteria and lactobacillus significantly,while the number of Enterobacter and Enterococcus reduced significantly.The results showed that probiotic beads have the metabolism and immune function regulating effects in mice with high fat diet,and have played lipid lowering role.This study paves new way for the use of probiotics-based products in improvement of human health.
