Purification,Modification And Moisturization Of Polysaccharides From Cortex Periplocae Of Different Producing Areas

The moistening property have a great impact on the aroma and smoking comfort of cigarettes.Traditional moistening agents have various advantages and disadvantages,and polysaccharide as a new moistening agent has better moistening effects.The Cortex Periplocae(CPP)has a variety of chemical components and biological activities,however,there is still lack of systematic research on the polysaccharide of it.In this paper,chemical composition detection,ultraviolet fullwavelength scanning,molecular weight determination,infrared spectroscopy,monosaccharide component determination,triple helix structure analysis were carried out on the purified polysaccharides of CPP from Sichuan,Hebei,and Henan province.The triple helix structure analysis,scanning electron microscopy(SEM),antioxidant activity,moisturizing application research and sensory quality evaluation of CPP modified by carboxymethylation and selenylation were carried out.The following conclusions were obtained:The water-soluble crude polysaccharides were extracted from Cortex Periplocae of Sichuan,Hebei and Henan using the "water extraction and alcoholic precipitation method",and a total of 12 polysaccharide components were obtained after separation and purification by DEAE-52 cellulose column,named SC-CPP0,SC-CPP1,SC-CPP2,and SC-CPP3(Sichuan province);HB-CPP0,HB-CPP1,HB-CPP2,and HB-CPP3(Hebei province);HN-CPP0,HN-CPP1,HN-CPP2,and HN-CPP3(Henan province).The UV full wavelength scan showed that these samples contained no or little protein and nucleic acid with high purity.The relative molecular weights of the 12 polysaccharides were 456,630,618,757,685,477,411,572,563,1035,1517,and 731 k Da,respectively,as determined by High performance liquid chromatography(HPLC),with good homogeneity of the samples.The IR spectra showed that all 12 polysaccharides contained characteristic absorption peaks of polysaccharides and contained uronic acid meant they were acidic polysaccharides,β-glycosidic bonds and furan rings.The results of the monosaccharide composition detected by HPLC showed that all CPP were heteropolysaccharides of different molar ratios composed of mannose,rhamnose,galacturonic acid,glucose,galactose,and arabinose,except for HN-CPP0,HN-CPP2,and HN-CPP3,which did not contain arabinose.Congo red experiments showed that SC-CPP0,SC-CPP2,SC-CPP3,HB-CPP0,HB-CPP3,HN-CPP0,HNCPP1,HN-CPP2 and HN-CPP3 have a triple helix structure,while SC-CPP1,HBCPP1 and HB-CPP2 do not have a triple helix structure.The carboxymethylation and selenization modifications of SC-CPP1,HB-CPP1 and HN-CPP2 were carried out and the IR spectra showed additional absorption peaks in the modified polysaccharide molecules near 1646,1385 cm-1,928,and 880 cm-1,respectively.The telescopic vibrational absorption peak of carboxymethyl and the symmetric variable angle vibrational absorption peak of saturated-CH as well as the Se-O-C and Se=O absorption peaks,demonstrating that the polysaccharide has been successfully modified by carboxymethylation and selenization.From the Congo red experiments on the CPP derivatives,it was clear that both the carboxymethylation and selenization modifications affected the triple helix structure of CPP from Sichuan and Hebei origins;but did not affect the triple helix structure of CPP from Henan.The main morphology of unmodified CPP from all three origins was smooth cylindrical granules of different sizes with dense structure and stacking phenomenon detected by SEM.After modification by carboxymethylation,the CPP showed a lamellar structure with a rough and porous surface.After the selenization modification,the CPP was formed by aggregation of spherical particles of different sizes.The analysis of the scavenging ability and reducing ability of CPP polysaccharides against DPPH radicals,ABTS radicals,OH radicals and O2-radicals showed that all CPP had certain antioxidant activity,with the CPP0 fraction of the three origins having the best antioxidant ability.The antioxidant capacity of SC-CPP1,HB-CPP1 and HN-CPP2 was enhanced by carboxymethylation and selenization modifications,with the antioxidant activity of carboxymethylated Cortex Periplocae polysaccharide being stronger than that of selenized Cortex Periplocae polysaccharide.SC-CPP1,SC-Ac-CPP1,and SC-Se-CPP1 were added to improve the moistening properties and sensory quality of cigarettes.The results showed that,SC-Ac-CPP1 and its derivatives could improve the moistening properties of cigarettes,and SC-AcCPP1 presented the best sensory evaluation with the addition of SC-Ac-CPP1 at 0.10%.
