Gloiopeltis furcata is one kind of most advantage algaes with economic value, it owns specifical pharmic and industrial values in application. The Gloiopeltis furcata with the founction of refreshing, digestion,anti-inflammation, eliminating damps contain a rich resource of agar,which is viscid. The antioxidation and anti-inflammation of Gloiopeltis furcata are connected with the presence of agar. The global increase in UVB radiation resulting from ozone depletion in the stratosphere, and the environmental degradation induce more and more complicated disease threatening human health. Searching safe and efficient UV radioprotectant, antioxidant and hygroscopic agent from food material with medicinal effect has been a focused issue.Mycosprine-like amino acids(MAAs) are naturally high-efficient ultra-violet absorbtion substances, it usually been found in cells of marine organism with the function of protecting aquatic organism from the hurt of ultraviolet. MAAs are water soluble substances characterized by a cyclohexenone conjugated with the nitrogen substituent of different amino acid, having small molecular weight, high molar extinction coefficient and absorption maxima ranging from 310 to 360 nm. The UV protection of MAAs is widely admitted, other activities such as antioxidation and anti-inflammation researches were reported in succession. There are MAAs in the cells of Gloiopeltis furcata, otherwise, algal polysaccharide has hydroscoicity and moisture retentive ability usually, but there is rare reports about the MAAs from Gloiopeltis furcata and other special active performance of MAAs from Chinese research scholars.This paper studied the extraction of MAAs from Gloiopeltis furcata harvest from Guangdong Province, including the species of MAAs and the optimizing extraction process of MAAs, and the oxidation resistance, the moisture-absorption and moisture-retention properties of MAAs from Gloiopeltis furcata. This paper is aimed to offer theoretical suggest for higher-value applying of Gloiopeltis furcata, also give suggest for MAAs applying in medicine and health, food, cosmetic, water-soluble paint and other industries. The main researches and results were as follows:1. The objective of this study was to explore the species of MAAs in Gloiopeltis furcata. Ultraviolet spectrophotometry, high performance liquid chromatography(HPLC), high resolution liquid chromatography-mass spectrometry technology(LC-MS) were used to analysis the methanol extract of Gloiopeltis furcata. The results indicated that methanol extract of Gloiopetis furcata showed the maximum absorption peak in 332 to 334 nanometer, and the high performance liquid chromatograph analysis indicated there were 5 kinds of MAA similar substance. Testing by LC-MS technology, the 5 kinds of substance in methanol extract of Gloiopeltis furcata were Shinorine, Mycosporine-2-glycine, Asterina-330, Porphyra-334, Deokygadusol, which were truly Mycosprine-like animo acids ingredients,the 98.3% of total content was Shinorine,which was the main ingredient.2. In order to maximize the extraction of MAAs components from Gloiopeltis furcata, we used the method of ultrasonic extraction with methanol as solvent, we determined the optimum range of ultrasonic temperature, ultrasonic time, liquid-solid ratio, methanol concentration, extraction times by a series of single factor experiments, then we optimized the extraction process of MAAs compounds from Gloiopeltis furcata by response surface methodology. On the basis of single factor experiment, the extraction condition of MAAs from Gloiopeltis furcata by ultrasonic extraction, optimized by Box-Behnken design, the results were ultraviolet temperature 41℃, extraction for 81 min,material/water ratio of 67mL/g, and methanol concentration 12%.Under this condition, the expected extraction rate of MAAs was 1.3115mg/g, and the actual extraction rate was 1.3064mg/g, while the extraction rate of traditional method was 0.9002mg/g, that is to say the extraction efficience of the optimized method can be 45.12% higher than the traditional method.3. The oxidation resistance test in vrtio for MAAs from Gloiopeltis furcata: The results showed the crude sample of MAAs from Gloiopeltis furcata played a good oxidation resistance in vitro, it possessed of a scavenging action to superoxide anion(·O2-)and hydroxyl radical(·OH), and the scavenging action showed a dose-dependent effect with the concentraction of the sample, the IC50(half maximal inhibitory concentration)to ·O2- and ·OH were 2.73mg/mL and 7.71mg/mL, the IC50 to DPPH was 13.679mg/mL, the clearance ratio to ·OH can be high to 90.45% when the concentration of MAAs from Gloiopeltis furcata, it is relatively stable in room temperature and light, this provide the theoretical basis for the further application of Gloiopeltis furcata.4. The moisture-retention properties of MAAs from Gloiopeltis furcata : The MAAs from Gloiopeltis furcata, glycerinum and hyaluronic acid were tested in vitro environment to measure their moisture-absorption and moisture-retention properties. The moisture-retention and absorption photometry of skin lotion and skin emulsion contained various concentration of MAAs from Gloiopetis furcata were also studied. The results indicated that the generally moisture-absorption and moisture-retention effects of MAAs from Gloiopeltis furcata were better than hyaluronic acid, but slightly lower than glycerinum. The MAAs from Gloiopetis furcata showed higher moisture retention efficiency than glycerinum and hyaluronic acid after 72 hours testing, it can retain water for a long time; The human skin testing indicated the skin lotion contained 60%MAAs concentrated solution showed the best moisture-retention efficiency, the skin emulsion contained 5% to 20%MAAs concentrated solution can significantly improved their absorbance value, the higher addition of MAAs concentrated solution, the higher absorbance value of skin emulsion.
