Nymphaea hybrid is one of large tropical lotus,containing various active ingredients and nutrients.The research on Nymphaea hybrid in China mainly focuses on the component analysis and the biological activity of alcohol extract,but few studies on the polysaccharide of Nymphaea hybrid.In the paper,the experiment used Nymphaea hybrid’s receptacle as raw material and mainly researched the extraction,separation and purification process,chemical and physical properties and moisturizing efficacy of Nymphaea hybrid polysaccharide,which improves the value of ornamental Nymphaea hybrid and provides the theoretical foundation for the application of Nymphaea hybrid polysaccharide as active ingredient in cosmetics.1.Nymphaea hybrid polysaccharide was obtained by water extraction-alcohol precipitation,based on the single factor experiments combined with BOX-Benhnken optimization analysis.The optimal conditions were to be liquid-to-solid ratio 35:1,extraction temperature 80℃,extraction duration 3h and the yield of the polysaccharide under above conditions could reach 12.073%.The optimal conditions of alcohol precipitation process were shown below: liquid-to-solid ratio 3.5:1,alcohol precipitation time 6h,ethanol concentration 90%.2.Activited carbon power,macroporous resin D315,H2O2 method was used for the decolorization of Nymphaea hybrid polysaccharide.H2O2 method were able to effectively decolorize Nymphaea hybrid polysaccharide and the decoloration rate was 80.5% at 340 nm.And three ways were adopted to remove protein from Nymphaea hybrid polysaccharide with Sevage method,trichloroacetic acid method(TCA),and TCA-Sevage combining method.The results showed that the percentage of protein removal was 70.49% and the percentage of polysaccharide loss was 29.24% with TCA method,which the best among the three methods.3.Nymphaea hybrid polysaccharide was purified sequentially by DEAE cellulose and Sephadex G-150 column chrom atographies,obtaining a major purified fraction named NHP-2.The molecular weight of NHP-2 was determined to be about romatography(HPLC)technology.NHP-2 was analyzed by infrared spectroscopy(IR)and nuclear magnetic resonance(NMR)spectrum.The results show that NHP-2 might be pectic substances and was made up of galacturonic acid.4.Compared with the traditional moisturizer,Nymphaea hybrid polysaccharide had good hygroscopicity and moisturizing performance with weighing and MTT method,and it can also promote keratinocytes to synthesize more AQP3 protein.Through the skin test of volunteers,it showed that Nymphaea hybrid polysaccharide had good moisturizing ability on the skin and provided a theoretical basis for the application of Nymphaea hybrid polysaccharide in cosmetics.
