Aging is an activity that accompanies the process of life occurrence and development and refers to the process of loss and degradation of the organism from constitutive substances and tissue structures to physiological functions.The social and economic burden is increasing with the increasing incidence of population aging and aging-related diseases.It is essential to functional research foods that delay aging and reduce the incidence of aging-related diseases to improve people’s living standards and quality of life.Chinese medicine has the characteristics of multi-component,multi-target,and multi-action mechanisms,which have obvious advantages in health care.Polygonati Rhizoma is rich in nutrients,plant polysaccharides,saponins,and flavonoids and has various functional activities such as antioxidants,aging delay,immunity regulation,and anti-fatigue.Zingiberis Rhizoma is a high-quality medicinal ginger unique to Sichuan,which has more warming and stimulating effects than the Zingiberis Rhizoma produced elsewhere,promotes blood circulation,and improves immunity.As a commonly used medicinal and food herb,Cinnamomi Cortex contains a variety of physiologically active substances such as cinnamon essential oil,oleoresin,flavonoids,polysaccharides,and polyphenols,which have high medicinal and edible value.Polygonati Rhizoma,Zingiberis Rhizoma,and Cinnamomi Cortex are medicinal and food ingredients,not only widely used in the field of medicine but also occupy an important position in the fiessentialalth food.As early as the Tang Dynasty,Sun Simiao’s"Thousand Gold Formula"used the combination of the three as a health cream to slow aging and improve beauty.However,there are few studies on the extraction process of the aqueous extract,anti-aging functional activity,and functional product development of the combination,and the application of the combination of the Mixed water extract of Polygonati Rhizoma-Zingiberis Rhizoma-Cinnamomi Cortex lacks theoretical basis and technical support.Therefore,in this paper,we select the high-quality raw material of Polygonati Rhizoma,optimize the extraction process,and study the in vitro antioxidant and in vivo anti-aging activity of the Mixed water extract of Polygonati Rhizoma-Zingiberis Rhizoma-Cinnamomi Cortex,and apply the Mixed water extract of Polygonati Rhizoma-Zingiberis Rhizoma-Cinnamomi Cortex to the research and development of anti-aging functional foods,to provide the theoretical basis and technical guarantee for its application in the field of functional foods.(1)By determining nutritional and active ingredients and in vitro antioxidant activity evaluation of different origins of Polygonati Rhizoma,it was found that Polygonati Rhizoma of Sichuan origin was more in line with the requirements of the subsequent experimental design and had certain advantages.(2)A response surface methodology was used to optimize the extraction process of the Mixed water extract of Polygonati Rhizoma-Zingiberis Rhizoma-Cinnamomi Cortex and the in vitro antioxidant activity of the Mixed water extract of Polygonati Rhizoma-Zingiberis Rhizoma-Cinnamomi Cortex was investigated.The in vitro antioxidant activity of the extracts was 0.37 mg/mL for DPPH,0.32 mg/mL,and ABTS~+0.48 mg/mL for superoxide anion radicals,indicating that they have good in vitro antioxidant activity.(3)Network pharmacology-based analysis of the anti-aging mechanism of action of the Mixed water extract of Polygonati Rhizoma-Zingiberis Rhizoma-Cinnamomi Cortex,the results showed that the neuroactive ligand-receptor interaction pathway,c AMP signaling pathway,diabetic complication AGE-RAGE signaling pathway,PI3K-Akt signaling pathway,and TNF signaling pathway might be related to the anti-aging functional activity of Polygonati Rhizoma-Zingiberis Rhizoma-Cinnamomi Cortex Combinations,and verified The feasibility of binding of key components Zingiberone,Shyobunone,Styrene,Alpha-Curcumene,quercetin,and beta-sitosterol with essential targets IL-1β,AGT,MAPK1,BCL2,TNF,MAPK3,NF-κB1,and IL-6 was verified.(4)The results of the effect of the Mixed water extract of Polygonati Rhizoma-Zingiberis Rhizoma-Cinnamomi Cortex on the antioxidant physiological function of aging mice revealed that the aqueous extract could reduce the levels of AGEs,IL-1β,and TNF-αand IL-6 in the liver tissues of aging mice,increase the organ indices of immune tissues liver,kidney,and spleen,increase the vitality of SOD,GSH-Px,and T-AOC and the content of anti-inflammatory factor IL-10 in liver and brain tissues,and reduce the NO vitality and MDA content in liver and brain tissues.(5)The analysis of intestinal flora biodiversity in aging mice revealed that the Mixed water extract of Polygonati Rhizoma-Zingiberis Rhizoma-Cinnamomi Cortex significantly increased the Chao1,PD,ACE and Shannon indices of the intestinal flora of aging mice increased the ratio of the number of Bacteroidetes to thick-walled Bacteroidetes in the intestinal flora of aging mice,decreased the relative abundance of Lactobacillus and Lactobacillus spp.and increased the relative abundance of Rumex.The functional prediction of intestinal flora revealed that the mass of microorganisms involved in metabolic pathways such as fatty acid metabolism and synthesis and bacterial infection in the intestinal flora of aging mice was significantly higher than that in normal mice and aging mice with the dietary intervention of Polygonati Rhizoma-Zingiberis Rhizoma-Cinnamomi Cortex mixture aqueous extract.(6)Based on the anti-aging activity of the mixed aqueous extract of Polygonati Rhizoma-Zingiberis Rhizoma-Cinnamomi Cortex,we developed a product with anti-aging functional activity-Polygonati Rhizoma-Zingiberis Rhizoma gelatin pastry,and the optimal formulation of Polygonati Rhizoma-Zingiberis Rhizoma gelatin pastry was optimized by orthogonal experiment:0.1%sugar powder,0.05%sodium alginate,4%maltodextrin and 1.25%maltose syrup based on the product yield.The quality of the product obtained from the optimal formulation was also evaluated regarding color difference,nutritional composition,and active ingredients.
