Rhizoma Polygonati which is perennial herb belongs to Polygona-turn Mill family. Modern pharmacological studies have shown that, Rhizoma Polygonati has many functions such as enhancing immunity, lowering blood sugar and blood fat, anti-aging, anti-virus, anti-tumor, anti-inflammatory, enhancing and improving memory and so on. This paper studied the contents of polysaccharides in Rhizoma Polygonati with different planting years which grew in Jiu Hua Mountain area in Anhui province; the author optimizied the parameters of the hot water extraction technology, also he studied the antioxidant activity of polysaccharides extrated from Rhizoma Polygonati. In addition, we studied the chemical constituents of essential oils in Rhizoma Polygonati and the scavenging effect of it on DPPH free radicals.Firstly, the author optimized the conditions of Phenol-H2SO4 colo-rimetry at 490 nm and the contents of polysaccharides in Rhizoma Polygonati of Jiu Hua Mountain was determined under the optimal conditions as follows, the amount of phenol 1.0ml, the amount of sulfuric acid 5.0 ml, color at 100℃for 15 min. Using the method of hot water extraction the author extracted the total polysaccharides in Rhizoma Polygonati of Jiu Hua Mountain including 1-10 years old, and the author determined its contents under the Phenol-H2SO4 colorimetry at 490 nm. The results shown that, with the extension of the growing period of Rhizoma Polygonati, the contents of polysaccharides generally increases; considering the various actual factors, the proper time for harvesting was in the 3 rd year after planting. Also, the results provided a theoretical basis for the standardized production of Rhizoma Polygonati in the area of Jiu Hua Mountain.Secondly, the author studied the hot water extraction of polysaccha-rides. The single factor experiments, orthogonal design have been used to optimize the conditions of extracting polysaccharides from Rhizoma Polygonati. The optimum hot water reflux extraction conditions of poly-saccharides from Rhizoma Polygonati of Jiu Hua Shan were obtained, temperature 100℃, ratio of solid-liquid 1:10, extraction time 2 h. Under these conditions, after 3 times of extraction, the yield of polysaccharides from powder of Rhizoma Polygonati was 15.99%; after 3 times of extra-ction, the yield of polysaccharides extracted from sheets of Rhizoma Polygonati was 11.65%. Meanwhile, we studied scavenging effect on hydroxyl radical of polysaccharides of Rhizoma Polygonati by Fenton reaction. The results showed that the ability of scavenging hydroxyl radicals of polysaccharides we purified was not obvious.Lastly, the essential oils in the fresh roots and stems of Rhizoma Polygonati were extracted by steam distillation and their constituents were identified by capillary gas chromatography-mass spectrometry. The results showed that,26 kinds of chemical components were identified in the roots, making up 90.87% of the total contents; 37 kinds of chemical components were identified in the stems, making up 94.58% of the total contents. In addition, the author studied scavenging effect on DPPH·of essential oils in the roots, the results showed that essential oils in the roots had a good scavenging effect on DPPH free radicals.
