| Constipation is a common intestinal disease in daily life.The incidence rate is on the rise worldwide,but people’s views on constipation have not risen to the level of disease.At the same time,the treatment methods of constipation also have certain drawbacks,which are prone to adverse effects reaction.In recent years,people are more and more inclined to prevent or improve the occurrence of constipation by eating natural foods.Camel milk has a variety of active ingredients,functional proteins and trace elements,which are excellent in anti-inflammatory,promoting digestion,and maintaining the stability of intestinal flora.Therefore,this study mainly explored the effect of camel milk on constipated mice,and provided theoretical support for camel milk intervention and prevention of constipation.The research contents are as follows:First,in this study,ICR mice were used as test subjects,and a constipation model was established by continuous gavage of loperamide hydrochloride solution for 7 days.The relieving effect of camel milk on constipated mice was verified by detecting changes in body weight,intestinal propulsion rate,time to the first black stool,and fecal water content.The results show that camel milk has preventive and ameliorating effects on loperamide hydrochloride-induced constipation mice,and can significantly relieve the basic symptoms caused by constipation.Secondly,the effect and mechanism of camel milk on constipation were explored by observing the pathological score and physiological index of colon tissue.The colon tissue of mice was observed by HE staining;the contents of gastrointestinal regulating peptides(MTL,Gas,SP,VIP,ET,SS)and serotonin(5-HT)were detected by ELISA test;AQP3and AQP4 were detected by real-time fluorescent quantitative PCR,C-kit gene expression;Western blot test detected AQP3,AQP4,C-kit protein expression.The results showed that camel milk could regulate the levels of constipation-related gastrointestinal regulatory peptides and 5-HT in the serum of mice to make them tend to normal levels,effectively reduce the protein and gene expressions of AQP3 and AQP4,and up-regulate C-kit.protein and gene expression.Finally,by examining the microbial diversity of mouse feces,To explore the effect of camel milk on the intestinal flora of mice,By observing the changes of mouse gut microbiota between different groups at alpha diversity,beta diversity,phylum level,genus level and species level.To determine the effect of camel milk on the intestinal microbial diversity of mice.The results showed that after induction with loperamide hydrochloride,the microbial diversity of mice decreased,and changes occurred within the group,Camel milk can effectively regulate the intestinal flora,improving the relative relationship between Proteus and Firmicutes at the phylum level,At the genus level,the relative abundances of Lactobacillus,Ligilactobacillus and Limosilactobacillus were increased,and the relative abundances of Acinetobacter,Muribaculum and Aerococcus were suppressed.In conclusion,camel milk has certain preventive and ameliorating effects on loperamide hydrochloride-induced constipation in mice,It can regulate intestinal flora and gastrointestinal regulatory peptides,protect the intestinal barrier,and inhibit the occurrence of constipation. |
