American ginseng berry is the mature berry of Panax quinquefolium L.,rich in ginsenoside,so it can be the stable raw material for the gain of ginsenosides.However,due to the limitation of technical conditions,there are still many shortcomings such as a single product,low utilization of raw materials as well as low value-added products during the production process of ginsenosides.Thus,it would be of extremely high commercial value and research significance to study the decolorization and refining process of ginsenosides from the American ginseng fruit.Our research focused on American ginseng berry,the results were shown as following:1?10 kinds of decolorizing materials?Activated carbon,magnesium oxide,calcium hydroxide,AB 8 macroporous resin,D301R macroporous resin,D280 macroporous resin,D900 macroporous resin,001×7 resin,HP-20 macroporous resin,and decoloring 1#macroporous resin?were selected to perform the decolorization experiment.By comprehensive comparison of retention and decolorization rates of the saponins,it was found that D301R macroporous resin had the best decolourization effect on the extract of American ginseng fruit while AB-8 was the most suitable macroporous resin for pigment recovery.The best decoloring conditions was determined to be:temperature 40?C,PH 5.30 and velocity1 BV/h.Under the best conditions,the decolorization rate was 84.26±1.56%,whereas the saponins retention rate was 88.75±1.86%.2?Preparation of pseudo-ginsenoside F11 and ginsenoside Re by using the“1-4-3”model simulated moving bed separation,the separation conditions were as follows:65%methanol was used as the elution solution,elution velocity was 60 mL/min,the residual liquid velocity was 30 mL/min,extracted pipe velocity was 30 mL/min,sample concentration was 2 mg/mL,switch step was 7 min.The purities of ginsenoside Re and pseudo-ginsenoside F11 were 91%and 93%respectively.3?Preparation of ginsenosides Rb2 and Rb3 by using the“1-4-3”model simulated moving bed separation,the separation conditions were as follows:70%methanol was used as the elution solution,elution velocity was 60 mL/min,the residual liquid velocity was 30 mL/min,extracted pipe velocity was 30 mL/min,sample concentration was 1.5 mg/mL,switch step was 11 min.The purities of ginsenosides Rb2 and Rb3 were 91%and 94%respectively.4?By using batch processing technology,making two C18 column??5*20cm?chained together,75%methanol as mobile phase,ginsenoside Rc and Rd were successfully prepared in one step with their purities of 89%and 91%respectively.5?Under sample concentration of 2 g/L,the 25%macroporous resin eluate of American ginseng berry saponins showed good insecticid activity towords Myaus persicae with 30%inhibition rate,while the 75%macroporous resin eluate of American ginseng berry saponins of displayed excellent repellent effect with 94.4%inhibition rate.Both pseudo-ginsenoside F11 and ginsenoside Re showed good repellent effect against Myaus persicae and Plutella xylostella,notably,the repellent effect of ginsenoside Re was almost 100%.In the DrosopHila melanogaster starving expriment,ginsenoside Re and pseudo-ginsenoside F11 showed obvious effect to female flies bred,the survival time could be extended to 4 days,and concentration of 1?M ginsenoside Re and F11 had the best effect.
