Croton laevigatus belongs to the genus of Croton(Euphorbiaceae). It is one of the traditional Chinese medicines.Domestic and international phytochemical studies on the plants of this genus indicated that there are abundant structural types with significant biological activities In order to search for the potential nematocidal active components of C.laevigatus.In this paper,we studied the chemical constituents and biological activities from the ethanol extract of C.laevigatus.Through tomographic techniques(silica gel,Sephadex LH-2,HPLC,TLC)and technology of spectrum(NMR,MS),twenty-one compounds(1–21)were isolated and identified from the ethyl acetate extract of the ethanol extract of C.laevigatus.Compounds1–3 were nor-sesqutierpenoids,4–8 were sesqutierpenoids,9–15 were diterpenoids,16–20were phenylpropanoids, and 21 were benzene derivatives. All of the compounds were isolated from C.laevigatus for the first time. Some compounds were tested for the biological activity(nematocidal activity, cytotoxic activities,anti-inflammatory activities.).The experimental results were observed by FDA/PI double-stained dot plots. The results showed that compounds 9–12,18,20 and 21 exhibited nematocidal activity with a lethality rate of over 60%,their lethality rates were 71.18%,86.44%,68.85%,67.58%,62.91%,91.42% and79.69%,respectively.Among them, compound 20 showed the highest lethality rate at91.42 %.All of the compounds didn’t show any cytotoxic activities to five human tumor cell lines or anti-inflammatory activities.Above all, this paper reported the isolation and identification of 21 compounds from the ethanol extracts of C.laevigatus, which expanded the chemical database. The nematocidal activity tests showed that diterpenoids and phenylpropanoids were some degree of nematocidal activity The result broadened the application of C.laevigatus and provided a reference for the research of new pesticides.
