| Styela plicata is an ascidian which belongs to class Ascidiacea,Subphylum Urochordata,Phylum Chordata.They mainly distribute north and south coastal in our country.According to the previous literature,the extraction of Styela plicata has significant biological activity,such as anti-tumor,anti-inflammatory,anti-virus,etc.It may has great potential value within the development of drugs.The research focused on isolation,structure identification and anticancer activities of the chemical constituents from styela tyela plicata,to provide a reference for the further construction of the marine natural product compound libraries and for the finding of lead compounds.It also has an important meaning to promote the efficient usage of marine resources.This paper mainly studied the organic solvents part of Styela plicata.The Styela plicata was extracted three times by 95%ethanol.Then the extract was partitioned by petroleum ether,ethyl acetate and n-butanol.Finally 20 chemica 1 constituents were isolated by silica gel column chromatography,Sephadex LH-20 gel column chromatography and HPLC methods.Based on the spectroscopi c data analysis,the chemical structures of 20 compounds were identified after comparised with the compound database literatures.The compounds’ isolated fr om Styela plicata were identified as two steroidal compounds,five nucleoside compounds,three butenolide compounds,three phthalic acid ester compounds,t wo organic acids,one pyranoside compounds and four other type compounds.Their structures were identified as Di-isobutyl phthalate(1),Di-n-butyl phthalate(2),Dodecanoic acid(3),1,2-benzenedicarboxylic acid bis(2S-methyl heptyl)est er(4),Palmitic acid(5),Cholest-5-en-3β-ol(6),(22E,24R)-ergosta-5,7,22-trien-3β-ol(7),5-hydroxy-3,4-dimethyl-5-pentylfuran-2(5H)-one(8),5-hydroxy-3,4-dimethyl-5-propylfuran-2(5H)-one(9),9-octadecenoic acid-2’,3’-dihydroxy propyl ester(10),ll-(5-hydroxy-3,4-dimethyl-2-oxo-2,5-dihydrofuran-5-yl)undecane acid ethyl est er(11),phenol,2,4-bis(1,1-dimethylethyi)(12),2’-deoxyuridine(13),2’-deoxythymid ine(14),(2E,6Z)-nonadien-1-ol(15),Ethyl-a-D-glucopyranoside(16),Uridine(17),Glycerol(18),Adenosine(19),2’-deoxyadenosine(20),respectively.All of them w ere isolated from styela plicata for the first time.11-(5-hydroxy-3,4-dimethyl-2-oxo-2,5-dihydrofuran-5-yl)undecane acid ethyl ester was identified as a new compound,so we evaluated the compound’s bioactivity of anti-tumor of A549 human lung adenocarcinoma cells,PC3 cells and MCF-7 breast cancer cells through the method of culturing cancer cells and CCK-8.The result showed that it could inhibit the proliferation of A549 human lung adenocarcinoma cells,PC3 cells,MCF-7 breast cancer cells at the concentration of 10-100 μg/mL,and the IC50 was 54.2,41.4,54.2 μg/mL,respectively. |
