BACKGROUNDThe customized and differentiated products gradually become dominant in the cosmetics market.Because of personalization and precision of 3D printing,the facial mask can be customized according to skin types,face shapes,and specific requirements of consumers,so as to improve the effect of the product.OBJECTIVEOur aim of this work was to take the anti-acne facial mask as an example to evaluate the feasibility and suitability of 3D printing facial mask.The hydrogel was selected and optimized in order to be suitable for 3D printing facial mask.And the quality,efficacy and safety evaluation of the 3D printing cryptotanshinone hydrogel mask were evaluated,which layed the foundation for the personalized and customized facial mask.METHODS1.Study on preparation of 3D printing cryptotanshinone hydrogel mask3D model of facial mask adapted to the physical characteristics was modeled by using 3D scanning and reverse engineering.The single factor and orthogonal test were used to optimize the hydrogel.The optimal formula,3D printing parameters and preparation process were determined,and 3D printing cryptotanshinone hydrogel mask was fabricated by 3D printing.2.The quality evaluation of 3D printing cryptotanshinone hydrogel maskThe quality of the 3D printing of the 3D printing cryptotanshinone hydrogel mask was preliminarily investigated according to the requirements of sensory and physicochemical indexes.SEM was used to observe the surface features of the 3D printing cryptotanshinone hydrogel mask.The method of the determination of cryptotanshinone concentration was established,and the stability of the functional component in the printing process was evaluated.In vitro release test was conducted by using vertical Franz diffusion cells to evaluate the release of the functional components in 3D printing cryptotanshinone hydrogel mask.3.Study on the pharmacodynamics of 3D printing cryptotanshinone hydrogel maskThe acne model of rats was induced by using oleic acid on the back skin of rats.After grouping and the treatment with 3D printing cryptotanshinone hydrogel mask,histological observation was used to observe the morphological changes of rat epidermis.The concentration of TNF-α and IL-8 in serum were determined and the expression of TNF-α and IL-8 in rats skin were detected by Western Blot test.4.The safety evaluation of 3D printing cryptotanshinone hydrogel maskThe skin safety of 3D printing cryptotanshinone hydrogel mask was evaluated by the rat skin irritation experiment and guinea pig skin allergy test according to the degrees of erythema and edema at the site of patch.And the pathological changes of rat and guinea pig epidermis were observed through the histological observation.RESULTS1.Study on preparation of 3D printing cryptotanshinone hydrogel mask3D model of facial mask adapted to the physical characteristics was modeled by using 3D scanning and reverse engineering.The optimal formula of hydrogel was optimized by the single factor and orthogonal test:sodium polyacrylate(NP-700)0.8 g,agar powder 0.6 g,glycerol 10.0 g.3D printing parameters and the preparation process of the 3D printing cryptotanshinone hydrogel mask were determined.2.The quality evaluation of 3D printing cryptotanshinone hydrogel maskThe sensory indexes and physicochemical indexes of the 3D printing cryptotanshinone hydrogel mask met the industry standard.The results of SEM characterization showed that the facial mask was homogeneous in texture and mechanical properties.The concentration of cryptotanshinone in the drug-loaded hydrogels before printing and the 3D printing cryptotanshinone hydrogel mask were respectively 99.79μg/g and 95.91μg/g,showing that no functional component degradation in the 3D printing process.In vitro release test results showed that the release rate of cryptotanshinone was consistent with the first-order equation.3.Study on the pharmacodynamics of 3D printing cryptotanshinone hydrogel maskHistological observation results showed that,after the treatment with 3D printing cryptotanshinone hydrogel mask,the thickening degree of stratified squamous epithelium in rats skin alleviated,hair follicle dilated slightly,keratinized cells decreased,dermal hyperemia and inflammatory cells decreased significantly,and the rats skin almost closed to the normal tissue.Histological classification results were statistically significant(P<0.05).Elisa test showed that the concentrations of TNF-α and IL-8 in serum decreased in different degrees,and the difference was statistically significant(P<0.01,P<0.05).Western Blot test showed that the expression of TNF-α and IL-8 in the skin was down-regulated compared with the model group.4.The safety evaluation of 3D printing cryptotanshinone hydrogel mask3D printing cryptotanshinone hydrogel mask showed no irritant reactions to rat skin and no allergic reaction to guinea pig skin.And there was no obvious pathological changes of rat and guinea pig epidermis by histological observation.CONCLUSION3D printing technology shows potential in the development of personalized facial mask.The optimized hydrogel is suitable for 3D printing facial mask,which displayed high drug loading.And there was no functional component degradation in 3D printing process.The quality of the 3D printing cryptotanshinone hydrogel mask was stable,and mechanical property was homogeneous.And cryptotanshinone could released from 3D printing cryptotanshinone hydrogel mask at the first order kinetic rate.The 3D printing cryptotanshinone hydrogel mask had anti-acne effect,and was safe when used externally.
