Sclerotinia sclerotiorum(Lib.)de Bary is an important plant fungal pathogen,and the sclerotinia stem rot(SSR)causes major economic and yield losses every year,which seriously restricts the development of oil crops in China.The pathogenesis of S.sclerotiorum is complex,and although it has been extensively studied,the mechanisms of its necrotrophic pathogenesis are still inadequately understood,and further research is needed in order to provide a reference for the prevention and treatment of SSR.S.sclerotiorum is a typical necrotrophic pathogen,plant hormone jasmonic acid plays key roles in plant resistance.Jasmonic acid signaling pathway plays an important role in the process of plant resistance to necrotrophic pathogens.Previously,we screened highexpression genes of S.sclerotiorum responding to jasmonic acid treatment through transcriptome data,and the data showed that SS1G_13659 was significantly up-expressed under jasmonic acid treatment.SS1G_13659 gene encodes an ABC transporter which has13 transmembrane domains and belongs to the ABCG subfamily.Split-Marker and homologous recombinant technology were used to knock out the SS1G_13659 gene,and two homozygous transformants ΔSS1G_13659-4 and ΔSS1G_13659-5 were obtained.The knockout of SS1G_13659 gene did not affect the hyphae growth,sclerotia formation,acid production and infection cushion formation of S.sclerotiorum,but the virulence of knockout transformants on the leaves of Arabidopsis thaliana and rapeseed was significantly reduced compared with that of wild-type strain.Interestingly,there was no significant difference in the virulence between the knockout transformants and wild-type strain on the jasmonic acid synthetic mutant opcl,suggesting that SS1G_13659 protein might be related to the resistance of S.sclerotiorum to jasmonic acid.The results will help us to further understand the pathogenesis of S.sclerotiorum and the role of jasmonic acid in plant resistance,and may provide new ideas and targets for the prevention and control of Sclerotinia disease.
