Objective To explore the expression of HNRNPC in cervical cancer by combining bioinformatics and immunohistochemistry,analyze the relationship between HNRNPC and the clinicopathological characteristics and survival prognosis of cervical cancer,and preliminarily explore the relationship between HNRNPC and the occurrence and development of cervical cancer,in order to find reliable molecular markers for the early diagnosis,prognosis assessment and precise treatment of cervical cancer.Method We select 60 patients including 20 cases of chronic cervicitis,20 cases of cervical intraepithelial lesions and 20 cases of cervical cancer by pathological examination in Affiliated Central Hospital of Shenyang Medical College from January 2000 to July 2020.Pathological and clinical date are complete.In this study,we use immunohistochemistry to detect the expression of HNRNPC in different cervical tissues.We use Continuity Corrected Chi-Square test to research the difference of expression levels in different pathological tissues.The expression difference of HNRNPC in normal cervical tissues and cervical cancer was preliminarily analyzed using several public tumor databases,such as TCGA,GEPIA,GEO,etc and the information in the database was further mined to analyze the relationship between HNRNPC expression and survival prognosis of patients.We use STRING database to construct the protein interaction model of HNRNPC and analysis the functional annotation.In order to explore the relationship between HNRNPC and immune cell infiltration,we use TIMER database.The influence of abnormal HNRNPC expression on tumor immune microenvironment was discussed.Result 1.The positive expression rate of HNRNPC in chronic cervicitis,cervical squamous intraepithelial neoplasia and cervical cancer tissues was 10%(2/20),20%(4/20)and 75%(15/20)respectively,the trend chi-square value was 18.262,P<0.001 was statistically significant.We use Continuity Corrected Chi-Square test to analyze the difference of expression levels in different pathological tissues.It was found that between chronic cervicitis group and SIL group there was no significant difference(P>0.05);The difference between the SIL group and the CC group was statistically significant(P<0.05);The positive expression rate of HNRNPC was statistically different between the chronic cervicitis group and CC group(P<0.05).2.We use Fisher accurately examined the relationship between HNRNPC expression and clinicopathological features of cervical cancer.In clinicopathologic features,HNRNPC expression was correlated with degree of cell differentiation(P=0.032).HNRNPC expression was not correlated with age(P=0.347),lymph node metastasis(P=1),and tumor stage(P=0.447).3.According to GEPIA database,HNRNPC is abnormally expressed in various cancers.4.According to TCGA database,HNRNPC is more highly expressed in cervical cancer tissues than that in normal cervical tissues.The results were statistically significant(P=0.01).5.Through the study of a set of clinical data of cervical cancer in the TCGA database,the results showed that high expression level of HNRNPC was significantly correlated with poor overall survival and disease-free survival of patients,that is,high expression of HNRNPC predicted the poor prognosis of cervical cancer patients,with statistical significance(P<0.05).6.The HNRNPC protein interaction model constructed by STRING database suggests that the 10 proteins with the most close interaction with HNRNPC are mainly involved in pre-m RNA processing,regulating m RNA cleavage,declination and translation.7.According to TIMER database,the expression level of HNRNPC was negatively correlated with the infiltration level of B lymphocytes(r=-0.178,P< 0.01)was statistically significant.ConclusionWith the deepening of cervical lesions,the positive expression rate of m6 A recognition protein HNRNPC increased,and the positive expression rate was significantly higher in cervical cancer.The expression of HNRNPC in cervical cancer is related to the degree of cell differentiation and poor survival.HNRNPC can be used for the early diagnosis of cervical cancer as a molecular marker,and can be used to evaluate the condition and prognosis of patients.
