The rapid propagation of the lianas like pueraria lobata and their adaptation in quarry slope are the key to achieve rapid vegetation coverage in quarry slope.In order to improve the survival rate of explants,callus induction rate,proliferation and differentiation efficiency,the rapid seedling growth system of pueraria lobata was optimized based on tissue culture technology.Through the observation of the growth index and photosynthetic index of tissue culture seedlings in the growth rule and suitability of the tissue culture seedlings in quarry slope were studied.Provide reference for the rapid breeding of pueraria lobata in north China and its field application in stone mines.The main results are as follows:1)In stem and leaf explants,the results of the study on the influence of different pretreatment and disinfection conditions on the survival rate showed that the best pretreatment method for stem was brush lightly,and the best disinfection conditions were75%C2H5OH treatment for 60s and 0.1%Hg Cl2for 8min.The best pretreatment method for leaf was soaking with detergent solution and then washing with water,and the best disinfection conditions were 75%C2H5OH treatment for 90s and 0.1%Hg Cl2for 8min.2)Studies on the effects of different plant hormones and concentrations on callus induction and proliferation showed that there was a great difference in callus induction rate when stem segment,stem tip and leaf were used as explants.The best explants for callus induction was stem tip and the optimal medium was MS+0.3mg/L6-BA+0.02mg/LNAA+0.3mg/L2,4-D,the optimal medium for callus induction was MS+1.5mg/L6-BA+0.5mg/LNAA.3)The research of the influence of different basic culture medium and plant hormones on callus differentiation show that with21QL medium as the basic culture medium can promote callus differentiation,relatively high concentrations of TDZ and GA3 on callus growth status has improved significantly,and in the process of differentiation KT and 6-BA mixture can effectively promote the callus differentiation.4)The study on the growth and photosynthetic index changes of pueraria lobata tissue culture seedlings applied in the quarry slope showed that,the growth indexes increased continuously during the observation period,and there was no significant difference between the growth of the quarry slope and the natural condition.The photosynthetic physiological indexes in each stage were positively correlated with air temperature and atmospheric CO2concentration.The comprehensive analysis of physiological indexes showed that pueraria lobata had strong drought resistance ability.It has strong adaptability in the quarry slope,and could be used in the ecological restoration of the quarry slope.Figure 34;Table 21;Reference 95...
