In recent years , the signal transmission in plants has been the researchfocus. In the progress of its growth , plant is influenced by all sorts of factsfrom in vivo and in vitro. Therefore,plant has to distinguish correctly andresponse relevantly, so as to guarantee healthy geowth and development.There are four stages of the molecular pathway in plant cell signaltransmission,which are , signal transmission between cells , signaltransmission on cell membrance , signal transmission in cells and proteinphosphorylation reversiblely. In advanced plant , abscisic acid (ABA)signal transmission take part in many important vital movement . Forexample , accelerating abscission , restraining growth , acceleratingdormancy , coursing stoma closure , regulating the development of seedsand embryo , affecting flowering , affecting gender differentiation .At the present time , the signal transmission mechanism of ABA ,which is a very important plant hormone , is known so little . In the knownfacts of ABA signal transmission , G protein or GTPase is the impressiveone . ROP GTPase (little G protein) regulates ABA signal transmission onmembrance . ROP (Rho-related GTPase from plants) is a unique branch ofRho GTPase in plant . More and more evidences prove that , the use of ROPprotein in plant is just like Ras, Rho and G protein in animal , is a kind ofcurrent sigal transmission switch . Their use in physiological anddevelopment are already analysed in transgene family , constitutively active,(CA), dominant negative, (DN), mutant . Researches prove that inArabidopsis thaliana and other species , ROP protein regulates cell polargrowth , pollen tube growth , cellular morphology building up , root hairgrowth and development , response from pressure of biological ornon-biological , response from many kinds of hormone (Auxin , Brassin ,ABA , etc ) . Remarkably , researches on obtain fuction and deletion fuctionsuggest that some ROP gene in Arabidopsis thaliana down regulates to ABAsignal transmission . Whatsmore , the localization in membrance is animportant condition of its fuction , which also proves the possibilty . In order to more deeply reseach the issues , which are ABA signaltransmission , mechanism and the part that G protein plays in the signaltransmission pathway , Arabidopsis thaliana wild type col-4 is taken asexperiment meteiral , and treated with ABA in different concentration .Then its ROPgefs and ROP10 gene which are related to ABA signaltansmission path are detected by Q-PCR . After that , the relationshipbetween ROPgefs and ROP10 protein is researched by Yeast twohybridation .The results of Q-PCR suggest that , the existance of ABA decreases thegene expression of ROPgefs .The gene expression of ROPgef1 is decreasedheavily . The gene expression of ROPgef13 is decreased lightly . The geneexpression of ROPgef14 is decreased a little . The gene expression ofROP10 is decreased in a small scale.According to Yeast two hybridation experiment , things are found thatthe relation between protein ROPgef1 and ROP10 is weak . Perhaps for thelocalization on the membrance , the relationship is weak . So the colorationshows negative . Whatsmore , some research is done on Pueraria lobata .The content of cellulose, hemi-cellulose, lignin and soluble saccharides aredetected , so as to provide referance to scientific basis for reasonableexploitation of Pueraria lobata resources . The result showed in the paperlaid the foundation for further study of ABA signal transmission pathwayand reasonable exploitation of Pueraria lobata resources .
