Male sterile of Arabidopsis are abundant and diverse such as morphological defects in flowers,defects in functional microspore,and anther non-dehiscence.Male sterile mutants may be good materials for the researchs about development of flowers,the microspore production,the pollens formation,anther dehiscence and gene function and so on.The mutants are beneficial to researching the male sterile mechanism,which lay a theoretical foundation for utilization of heterosis in rapeseed.In our study,we obtained non-petals mutants and non-dehiscence mutants of Arabidopsis.The research about preliminary phenotypic analysis and genetic analysis in mutants.And elucidated the mechanism about ap3-like,the 55 gene was isolated about dehiscence,and the anther non-dehiscence was related to DNA methylation.The major results were described as follows:1.There were some morphological defects in flowers of ap3-like mutant,including lacks of stamens and petals which were similar to that of ap3-3 mutant.There was no change in vegetative growth and other character of mutant comparing with wild-type.2.In mutant of rapeseed showed some defects,such as wizened and smaller petals,stamens which produce lesser pollens.Scanning electron micrograph showed that the petals had irregularly shaped but elongated epidermal cells and stomas,while the cells in wild plant flowers were well organized and closely arranged.3.The transgenic research suggested that it was not the transgenic construct was responsible for the ap3-like phenotype.4.The molecular biology and genetics analysis showed that it was the AP3 promoter methylation responsible for the low expression of the AP3 and the ap3-like phenotype.5.In the complementation lines the AP3 promoter methylation was decreased to 20%,and the expression level of AP3 was rescued.6.The M6 and M24 mutant showed male sterile,and the Alexander’s staining of pollen showed that the pollens were the same as wild-type pollen.And there was no pollen outside the anther,the scanning electron micrograph showed the anther of mutant was indiscent.7.The genetics analysis showed that the phenotype of the anther non-dehiscence was comtrolled by one gene.8.In the genetics analysis using the mutants about epigentics to build double mutants and suggested that the anther non-dehiscence was related to the DNA methylation.9.Using the genome wide methylation analysis looking for genes.And genetics analysis and transgenes research were submitted to the function of the genes,and showed the 55 gene may be responsible for the anther dehiscence.
