As a common fruit in people's daily life,apples are rich in minerals and vitamins,so they are deeply loved by everyone.As China's important apple producer and consumer country,China occupies an important position in the world's apple industry.Shaanxi is one of the main apple producing areas in my country.In the past 20 years,the apple industry has contributed to increasing the income of local farmers and farmers.Made a huge contribution to getting rid of poverty and becoming rich.With the continuous expansion of the local apple industry,the quality and yield of apples have gradually attracted the attention of all walks of life and farmers.People have found that the quality and yield of apples are not only related to the genetics of the fruit tree,but also closely related to the rhizosphere microorganisms.This article mainly focuses on the research of different ages of apple trees in Yan'an area and the correlation of rhizosphere microorganisms,hoping to provide a certain theoretical basis and reference value for improving the quality and yield of apple trees.The main research results are as follows:(1)The results of the physical and chemical properties of the rhizosphere soil of apple trees of different ages showed that the ammonium nitrogen content of apple trees of different ages in the same area gradually decreased with the increase of tree age;among them,the ammonium nitrogen content of LC7 was the highest,reaching 34.00 mg/kg;30a apple tree rhizosphere soil contains the highest content of available phosphorus(72.00 mg/kg),while the available phosphorus content of apple tree rhizosphere soil in Yan'an is generally higher than that in Luochuan County.(2)The high-throughput results showed that 6 samples were spliced into 1670 OTUs on average.The petal image showed that 6 samples had a total of 625 OTUs,while the number of OTUs unique to LC7,LC15,LC20,LC24,LZ30 and LG50 were 54,21,82,42,120,71.Analyzing the diversity and richness of the microbial communities in the samples found that LZ30 and LG50 have higher species diversity,while LC20 has the highest microbial community richness.(3)Spearman correlation coefficient and db RDA analysis were used to study the correlation between physical and chemical factors and microbial communities.At the phylum level,there was a significant positive correlation between the content of AP and Tenericutes soft-walled bacteria(P <0.05),and a significant negative correlation with Cyanobacteria(P <0.05).There was a very significant positive correlation between the content of available potassium(AK)and the phylum Elusimicrobia(P <0.01).At the genus level,there was a significant positive correlation between the content of ammonium nitrogen and Solirubrobacter(P <0.05);the content of available phosphorus(AP)and Steroidobacter was extremely significantly positively correlated(P <0.01);the content of available potassium(AK)and Nocadioides There was a significant positive correlation between Nocardia(P <0.05).(4)The Illumina Mi Seq high-throughput sequencing technology was used to analyze the diversity of rhizosphere soil bacteria in different habitats in Yan'an and different yields of apple trees in the same habitat,and a large amount of relatively comprehensive bacterial community information was obtained.A total of 591,413 effective sequences were generated from 8 samples through high-throughput sequencing,with an average of 73,927 effective sequences per sample,and a total of 13,688 OTUs were generated with 97% agreement.(5)The results of CCA analysis in the correlation analysis of environmental factors indicate that bacterial diversity is mainly affected by the content of available phosphorus in the soil,which indicates that the content of available phosphorus in the soil has a certain impact on bacterial diversity,followed by soil p H,ammonium nitrogen,available potassium and The content of organic matter.
