Research Background:Pancreatic cancer is a frequent cancer in the digestive system.People over 40 years old are vulnerable,male incidence is greater than female.The five-year survival rate was only 1.2%-6%and the median survival time was 21.1 months.In recent years,the incidence of pancreatic cancer has increased.There is a lack of specific clinical manifestations in the early stage of pancreatic cancer.Many patients have reached the stage of advanced or even distant metastasis when diagnosing pancreatic cancer,and only 10%-20%of pancreatic cancer patients are feasible for surgical resection.The diagnosis of pancreatic cancer is still mainly based on clinical manifestations,features and imaging results at present.Therefore,it is urgent to find more specific and sensitive diagnostic techniques to improve the early diagnosis rate of pancreatic cancer.Objective:To investigate the expression and significance of MMP-12(matrix metalloproteinase-12)and B7-H3(costimulatory molecule B7 family homologue 3)in pancreatic cancer,and to explore the relationship between them and clinicopathological parameters of pancreatic cancer.Methods:45 cases of pancreatic cancer tissues and corresponding adjacent tissues were collected from November 2017 to April 2019 in the Department of general surgery of our hospital.The expression levels of MMP12 and B7-H3 were detected in pancreatic cancer tissues and their corresponding paired adjacent tissues by immunohistochemistry,their relationship with clinicopathological parameters were analyzed,and the correlation of MMP-12 and B7-H3 expression in pancreatic cancer tissues was further analyzed.Results:1.The positive expression rate of MMP12 in pancreatic cancer was more higher than that in paired paracancerous tissues(44.4%vs7%,P<0.05).The expression level of MMP12 in pancreatic cancer tissues showed a statistical difference when compared with the data of pancreatic cancer with or without perineural invasion and the clinical TNM stage,while no significant correlation was found in parameters such as age,gender,tumor location,tumor size,tumor stage,differentiation grade,and the presence or absence of lymph node metastasis.2.The positive expression rate of B7-H3 in pancreatic cancer tissue was much higher than that in paired paracancerous tissues(71.1%vs15.5%,P<0.05).The expression levels of B7-H3 in pancreatic cancer tissues were not significantly correlated with clinicopathological parameters of pancreatic cancer patients,such as age,gender,tumor location,tumor size,clinical stage,differentiation,presence or absence of lymph node metastasis,and presence or absence of perineural invasion.3.There was a positive correlation between MMP12 and B7-H3 protein expression in pancreatic cancer(R2=0.484,correlation coefficient 0.697,P R<0.05)Conclusions:MMP-12 and B7-H3 showed higher expression levels in pancreatic cancer tissues compared with paired paracancerous tissues,and There was a significant positive correlation between MMP-12 and B7-H3 expression in pancreatic cancer tissues.MMP-12 and B7-H3 maybe combined as diagnostic markers of pancreatic cancer.
