Objective: To study the mechanism of acupuncture in modulating chronic inflammation and IR via SIRT1.The project will disclose the important mechanism of acupuncture in improving IR and chronic inflammation,providing new proof for clinical application of obesity.Methods: 40 Wistar rats(8 weeks old,male)were randomly divided into four groups including normal group,obesity group,EA group and resveratrol group.Normal group received normal laboratory diet while others received high-fat diet continued throughout the experimental process(16 weeks).EA and resveratrol treatment carried out from the 8th week to the 16 th week,three times each week.Weight and food intake were analyzed every two weeks after acclimation.Fasting blood glucose(FBG)and postprandial blood sugar(PBG)were measured every two weeks during treatment.Intraperitoneal insulin tolerance tests(IPIT)and intraperitoneal glucose tolerance tests(IPGT)were performed after six weeks of treatment.Serum FFA,triglyceride(TG)and total cholesterol(TC)were analyzed after treatment.Rats were sacrificed at the end of the treatment and excised WAT for protein and RNA extraction.The protein or gene expression of Sirt1,NF-?B,and TNF-? were tested though Western Blotting or Real-Time Reverse Transcriptional Polymerase Chain Reaction.Meanwhile,the types of macrophages were analysed also.Results: There were no statistical differences in all groups at food intake,FBG and PBG.High fat diet increased body mass,TG,TC and FFA levels,and also reduced insulin sensitivity and glucose intolerance which was established through IPIT and IPGT compared with normal group.Electroacupuncture(EA)and resveratrol treatment reduced body mass,TG,TC and FFA levels,and also improved insulin sensitivity and glucose intolerance which was established through IPIT and IPGT in IR obesity rats.It was likewise founded EA and resveratrol activated Sirt1,which could deacetylate NF-?B,down-regulating the expression of TNF-?.Macrophage 1 was critical in the process of EA or resveratrol used to treat obesity also.Conclusions: IR obesity rat model was successfully induced with high fat diet and results indicated that EA offers a beneficial effect on insulin resistance and inflammation in IR obesity rats through Sirt1/NF-?B/TNF-? signaling.
