Purpose: An experimental study of acupuncture combined with moxibustion to improve aortic remodeling in rats with normal high blood pressure by regulating miR-126.Material and method:A total of 48 experimental rats were selected,and 40 healthy male Dahl salt-sensitive rats(DS)were divided into 5 groups by random number table method,with 8 rats in each group,which were model group,acupuncture group,acupuncture + moxibustion group,non-channel and non-acupoint group.In blank group,8 healthy male salt-resistant rats(SS)were fed with 8%Na Cl high salt diet to establish normal high blood pressure model.Do not take any measures to blank group and model group,acupuncture group received electroacupuncture "st 36","li 11",moxibustion stick stick moxibustion group stick to "spleen Shu",acupuncture plus moxibustion groups treatment of acupuncture and moxibustion posts set of joint application,without going through the acupuncture point group(15 mm on bilateral iliac crest,next to the midline open after 20 mm)accept electroacupuncture,treatment time and the same as the acupuncture group,a total of 4 weeks treatment.After treatment,arterial blood of rats was collected and serum was separated.The contents of ET-1,NO,SOD and MDA in serum of rats were detected by ELISA.The tissue of thoracic aorta of rats was collected,and the morphological changes were observed by HE staining after fixation.The protein expressions of MMP-2,MMP-9 and AngⅡ in thoracic aorta were detected by Western Blot.The mRNA expression of MMP-2,MMP-9 and AngⅡ in thoracic aorta was detected by reverse transcription polymerase chain reaction(RT-PCR).The expression of miR-126-3p/5p in thoracic aorta was detected by real-time fluorescence quantitative PCR(qPCR).Results:1.Acupuncture + moxibustion therapy can regulate blood pressure in rats with high normal blood pressure.The experimental results show that 1 week after treatment,compared with model group,acupuncture group,moxibustion stick,without going through the acupuncture point group,acupuncture plus moxibustion stick rat tail artery systolic blood pressure is reduced,there is statistical significance difference(P < 0.05),and the group of acupuncture,moxibustion stick and compared without going through the acupuncture point group,acupuncture plus moxibustion stick group rat tail artery systolic blood pressure reduced more significantly(P <0.05);After 2 to 4 weeks of treatment,the systolic blood pressure of the caudal artery of rats in the treatment groups showed a continuous decreasing trend,and there was a statistical difference compared with the model group(P < 0.05),and the decreasing degree of the acupuncture + moxibustion group was significantly better than that of the other groups(P < 0.05).After 2 to 4 weeks of treatment,the systolic blood pressure of the caudal artery in each group was stable,and there was no significant difference compared with 2 weeks of treatment(P BBB 0 0.05).2.Acupuncture and moxibustion can improve the morphology of thoracic aorta in rats with high normal blood pressure.The results of HE staining showed that the intima of the model group was obviously thickened,smooth muscle cells were arranged disorderly,and foam cells were infiltrated.The intima of the acupuncture group,the moxibustion group and the non-meridian and non-acupoint group were slightly thickened,the endothelial cells of the tube wall were more continuous,the medial membrane was thinner than that of the model group,and the degree of hyperplasia was decreased.However,in the acupuncture + moxibustion group,the intimal hyperplasia of aorta was not obvious,and there was no significant difference in the thickness between the internal and external membrane and the blank group.Compared with the acupuncture group,the moxibustion group,the non-meridian and non-acupoint group and the model group,the morphological improvement of the damaged tissue was more significant.3.Acupuncture and moxibustion can regulate oxidative stress in rats with high normal blood pressure.Acupuncture and moxibustion can regulate the level of oxidative stress in rats with normal high blood pressure.Elisa results showed that the serum NO、SOD level of the model group was low and the ET-1、MDA expression was high.after treatment,the serum NO、SOD level of each treatment group increased in varying degrees,and the ET-1、MDA expression decreased statistically(P<0.05),while the serum NO、SOD level increased and the ET-1、MDA expression decreased more significantly in acupuncture moxibustion group than in acupuncture group and moxibustion group.4.Acupuncture and moxibustion can improve the gene expression of extracellular matrix metalloproteinases in rats with normal high blood pressure.RT-PCR results showed that compared with the blank group,the mRNA levels of AngⅡ,MMP-2 and MMP-9 in the model group were up-regulated(P<0.05).After treatment,the mRNA expressions of AngⅡ,MMP-2 and MMP-9 in the treatment groups were down-regulated(P<0.05),and the mRNA expressions of AngⅡ,MMP-2and MMP-9 in the acupuncture + moxibustion group were down-regulated more significantly,with statistical significance(P<0.05).5.Acupuncture and moxibustion can improve the protein expression of extracellular matrix metalloproteinases in rats with high normal blood pressure.WB results showed that compared with blank group,the protein expression levels of AngⅡ,MMP-2 and MMP-9 in model group were higher(P<0.05).After treatment,the protein expressions of AngⅡ,MMP-2 and MMP-9 in each treatment group were down-regulated(P<0.05),and the protein expressions of AngⅡ,MMP-2 and MMP-9in the acupuncture + moxibustion group were down-regulated more significantly,with statistical significance(P<0.05).6.Acupuncture and moxibustion can regulate miR-126 to induce its target gene to improve thoracic aortic remoremoing in rats with normal high blood pressure.qPCR results showed that compared with the blank group,the expression miR-126-3p、miR-126-5p thoracic aorta in the model group was significantly decreased(P<0.05).The miR-126-3p、miR-126-5p expression increased in different treatment groups(P<0.05),and the miR-126-3p、miR-126-5p expression in acupuncture and moxibustion group was higher than that in other groups.Conclusion:1.acupuncture combined with moxibustion paste can improve the blood pressure of rats with normal high blood pressure,which is more significant than that of acupuncture and moxibustion alone.2.acupuncture combined with moxibustion can effectively regulate blood pressure in rats with normal high blood pressure by regulating serum ET-1、MDA、NO、SOD level,improving oxidative stress state,regulating vascular endothelial function and free radical level,and scavenging vascular inflammatory reaction.3.acupuncture combined with moxibustion can down-regulate the mRNA and protein expression of thoracic aortic tissue AngⅡ、MMP-2、MMP-9 and improve the process of vascular remodeling.4.acupuncture combined with moxibustion and paste can regulate miR-126,vascular endothelial function,protect damaged blood vessels and improve aortic remodeling to regulate blood pressure in rats with normal high blood pressure.
