Objective: Chronic fatigue syndrome(CFS)has become one of themain forms ofsub-health and has potential harm to human beings.With the increase of social pressure and the increase of sub-health people,the development of oral anti-fatigue agents has become the need of health,society and market.At present,many anti-fatigue preparations have been made into various dosage forms,but the development prospect is still broad.In this paper,astaxanthin with good antioxidant activity and Lijia capsule from Tianjin Tasly Pharmaceutical Co.,Ltd.were selected as anti-fatigue preparations to determine their effects on anti-chronic exercise fatigue,so as to provide new possibilities for anti-chronic exercise fatigue.Materials and methods: Select 30 only 4 weeks of male SD rats as experimental animals,each group 5,divided into astaxanthin motility chronic fatigue group(A),the better capsule motility(L),chronic fatigue group contrast motility chronic fatigue group(C),astaxanthin motility chronic fatigue exhausted group(EA),the better capsule motility chronic fatigue exhausted group(EL),The control group was chronic fatigue exhaustion(EC).The chronic exercise-induced fatigue model is divided into two stages: adaptive training and strengthening training,with a period of 60 days.In the first 30 days,the rats ran for three days at a speed of 10m/min and rested for one day,so that the rats were fully adapted to running training.Then the training was intensified.From the 32 nd day to the 34 th day,the exercise speed was 15m/min and the exercise duration was 20 min.On the 36 th to 38 th day,the exercise speed was20m/min,and the exercise time was 20 min.From the 40 th day to the 42 nd day,the exercise speed was 27m/min and the exercise time was 20 min.From day 44 to day 46,the exercise speed was 30m/min,and the exercise time was 20 min.From day 48 to day50,the exercise speed was 35m/min,and the exercise time was 20 min.From day 53 to day 55,the movement speed was 37m/min,and the movement time was 30 min.From day 56 to day 57,the exercise speed was 39m/min and the exercise time was35 min.From the 58 th day to the 59 th day,the exercise speed was 40m/min and the exercise time was 40 min.At the same time of modeling,the experimental animals were given astaxanthin intervention dose of 60mg/kg/d,Lijia capsule intervention dose of 30mg/kg/d,the volume of cooking oil was 1 m L,and the placebo group was given cooking oil of 1 m L/day.At the same time,exercise training was carried out daily according to the chronic exercise fatigue regimen.In the exercise-induced chronic fatigue group,serum,liver tissue and gastrocnemius mitochondria were collected 24 hours after the last exercise.In the exercise-induced chronic fatigue exhaustion group,a high-intensity exhaustive exercise was performed 24 hours after the last exercise training,and the exhaustive time was recorded.The mitochondria of gastrocnemius muscle were extracted,the respiratory control rate was measured by Clerk electrode method,and the mitochondrial membrane potential was measured by JC-1 method.Serum was collected and the contents of BUN,LDH,NEFA,CK and glycogen were determined by ELISA.The levels of antioxidant enzymes CAT and GSH-Px were determined by ELISA.The ratio of GSH/GSSH was determined by ELISA.The expression levels of TNF-? and NF-?B were determined by Western Blot.The level of MPO in tissues was determined by immunohistochemistry.Results:1.Anti-fatigue effect of astaxanthinAfter intervention of astaxanthin,compared with control group,the motility of chronic fatigue group rats running exhaustion time increased significantly(p < 0.05),BUN content decreased significantly(p < 0.05),NEFA content increased significantly(p < 0.05),liver glycogen content increased significantly(p < 0.05),CK content decreased significantly(p < 0.05),suggesting astaxanthin can improve the tolerance of rats to exercise intensity,improve the ability of function,increase the movement time;After astaxanthin intervention,compared with the control group,the content of GSH-Px was significantly increased(P < 0.05),the activity of CAT enzyme was significantly increased(P < 0.05),and the GSH/GSSG was significantly increased(P< 0.05),indicating that astaxanthin can enhance the antioxidant level of rats and reduce the harm of oxidative damage.After astaxanthin intervention,compared with control group,the mitochondria of rats MMPS increased significantly(p < 0.05),mitochondrial RCR increased significantly(p < 0.05),after intervention of astaxanthin,rats are enhanced mitochondrial function,can work well rats did not provide energy,can play the role of improve the sport ability of rats;Western Blot results showed that after astaxanthin intervention,TNF-? in astaxanthin intervention group was significantly decreased compared with the control group(P < 0.05),indicating that astaxanthin can reduce the production of inflammatory cytokines and protect tissues from injury.The results of MPO immunohistochemistry showed that the MPO of astaxanthin intervention group was lower than that of control group,indicating that astaxanthin can reduce the production of inflammatory cytokines.2.Anti-fatigue effect of Lijia capsuleAfter the better capsule intervention,compared with the control group,chronic fatigue group running at the time of the rats increased significantly(p < 0.05),BUN content decreased significantly(p < 0.05),NEFA content increased significantly(p < 0.05),liver glycogen content increased significantly(p < 0.05),CK content decreased significantly(p < 0.05),explain the beautiful capsules can improve the tolerance of rats to exercise intensity,improve the ability of function,increase the movement time;After Lijia capsule intervention,compared with the control group,the content of GSH-Px was significantly increased(P < 0.05),the activity of CAT enzyme was significantly increased(P < 0.05),and the GSH/GSSG was significantly increased(P< 0.05),indicating that Lijia capsule can enhance the antioxidant level of rats and reduce the harm of oxidative damage;After the capsule intervention,compared with control group,the mitochondria of rats MMPS increased significantly(p < 0.05),mitochondrial RCR increased significantly(p < 0.05),shows that after the capsule intervention,rats are enhanced mitochondrial function,can work well rats did not provide energy,can play the role of improve the sport ability of rats;MPO immunohistochemical results showed that the MPO of astaxanthin intervention group was lower than that of control group,indicating that Lijia capsule can reduce the production of inflammatory factors and protect tissues from damage.Conclusion:The exercise ability of the rats was enhanced with the intervention of astaxanthin or Ritka capsules.These two preparations can achieve the effect of anti-chronic exercise fatigue through anti-oxidative stress,anti-inflammation and other pathways.
