Objective: exercise fatigue is a normal physiological phenomenon in the process of human movement,but if it is not eliminated in time,it will produce excessive fatigue,and long-term accumulation will harm health.With the upsurge of national fitness,it is more and more common to take exogenous supplements to relieve fatigue.Therefore,in this study,the mice with acute exercise fatigue were taken as the experimental objects,and the exogenous intake of astaxanthin or Lijia capsule was taken as the intervention method.The biochemical level,inflammatory level and mitochondrial function were discussed to observe the effect of astaxanthin and Lijia Capsule on alleviating fatigue,so as to reduce the skeletal muscle injury caused by acute high-intensity exercise and protect the cell structure,it provides new solutions and ideas to relieve exercise fatigue.Methods: Thirty-six Kunming male mice of 2 months old were randomly divided into 6 groups,6 in each group: exhaustive exercise control group(C+E),astaxanthin exhaustive exercise group(AX+E),Lijia capsule exhaustive exercise group(L+E),quantitative acute exercise control group(C+AE),astaxanthin pre scheduled acute exercise group(AX+AE),Lijia capsule intervention quantitative acute exercise group(L+AE).The way of drug intake in mice was gavage.The intervention dose of astaxanthin was 3mg/100g/day,the intervention dose of Lijia capsule was600mg/100g/day,and the placebo group was olive oil,the dose was 0.1 ml/100g/day.After 6 weeks of drug intervention,mice were given one exhaustive exercise and quantitative acute exercise respectively.The exhaustive exercise program was: mice ran on the treadmill.The initial velocity is10m/min,5°After that,the running speed was increased by 3m/min every 3 min until exhaustion.At the end of exercise,the mice showed weakness,weakness,unable to adhere to the original running speed,ineffective driving by external electrical and human stimulation,faster respiratory rate after stopping exercise,body lying on the ground,slow response after external stimulation,which was the exhaustion standard;The quantitative acute exercise program was: mice ran on the treadmill.5° the initial speed was 10 m/min,then increased by 3 m/ min every 3 min after 5 min,and the total exercise time was 25 min.Three hours after the end of exercise,blood was taken from the orbit of the mice.After the blood was taken,the mice were killed and the skeletal muscle was taken for standby.The levels of urea nitrogen,free fatty acid,lactate dehydrogenase and liver glycogen in serum were determined by ELISA to indicate fatigue level;The mitochondrial oxidative phosphorylation function of gastrocnemius muscle was determined by Clark electrode method;He staining was used to observe the inflammation and injury;The protein of skeletal muscle was extracted,and the inflammatory related factors IL-1,COX2,IL-6 and TNF were determined by Western blot-? The expression level of the protein was higher.The results are as follows:1.Effect of astaxanthin on exhaustive exerciseAfter 3 hours of exhaustive exercise,compared with the control group,the contents of bun and LG in astaxanthin intake group increased significantly(P<0.05),and the activity of LDH decreased significantly(P<0.05);In astaxanthin group,mitochondrial RCR increased(P<0.05)and mitochondrial ATP concentration increased significantly(P<0.05),indicating that astaxanthin intake increased mitochondrial energy production efficiency;Morphological observation showed that the skeletal muscle cells in astaxanthin intake group were more regular in shape,closely arranged and clear in outline than those in the control group.The muscle cells in cross section were intact,without obvious swelling,with less blurred shape and less inflammatory cell invasion,indicating that astaxanthin can reduce gastrocnemius injury and inflammation;Western blot showed that the expression of IL-6 protein in astaxanthin treated group was significantly higher than that in control group(P<0.05),TNF-? the expression level was lower than that of the control group(P<0.05);The changes of IL-1 and COX2 were not statistically significant.2 Effect of astaxanthin on fatigue recoveryFor the quantitative acute exercise group,compared with the control group,the bun content and LDH activity of astaxanthin intake group were significantly increased(P<0.001)and decreased(P<0.05),indicating that astaxanthin intake can reduce the degree of skeletal muscle injury;Compared with the control group,the mitochondrial RCR ratio and ATP concentration of astaxanthin intake group were significantly increased(P<0.05),indicating that astaxanthin intake increased the mitochondrial energy supply effect during quantitative acute exercise;Morphological observation showed that compared with the control group,the gap of skeletal muscle cells in astaxanthin intake group was smaller,the cell area was more complete,and there was no large number of inflammatory cells infiltration.3 Effect of Lijia Capsule on exhaustive exerciseCompared with the control group,the content of BUN in the intervention group increased significantly(P<0.05)and the activity of LDH decreased significantly(P<0.05),indicating that the intake of Lijia capsule can promote the alleviation of fatigue;The RCR ratio and MMP level of gastrocnemius mitochondria in Lijia capsule group were significantly different from those in the control group(P<0.05);Morphological observation showed that the inflammation of Lijia capsule group was not significant,the cells arranged closely,the shape was regular,the inflammatory area was small,and the number of nuclei was large,suggesting that Lijia capsule can reduce gastrocnemius injury and inflammation;Western blot results showed that the levels of IL-1,COX 2 and TNF in Lijia capsule intervention group were significantly higher than those in control group after 3 hours of exhaustion recovery-? The expression level was significantly lower than that of the control group(P<0.05).4 Effect of Lijia Capsule on fatigue recoveryIn Lijia capsule intervention group,after 3 hours of acute quantitative exercise recovery,the level of MMP in gastrocnemius mitochondria was significantly increased(P<0.05),and the concentration of ATP was significantly increased(P<0.05).Conclusion: astaxanthin or Lijia capsule can relieve acute exercise-induced fatigue in mice.Astaxanthin can prolong the exercise time of mice,delay the occurrence of exercise fatigue,accelerate the recovery of mitochondrial respiratory function of skeletal muscle after exhaustive exercise,increase the level of IL-6 in skeletal muscle,and reduce the level of TNF-? in skeletal muscle to eliminate inflammation and promote fatigue recovery;Lijia capsule can improve mitochondrial membrane potential,increase intracellular ATP concentration and protect mitochondrial function.It can also reduce IL-1,COX2 and TNF-? Expression level,protect skeletal muscle from inflammatory damage,has a better effect on the protection of skeletal muscle injury.
