BackgroundPsoriasis is a chronic disease.In the acute stage,blood heat is the first,and latent heat is inherent.When the heat is excessive,the body fluid is injured,or the treatment is improper,or the time is prolonged,the Yin fluid will be consumed,the skin will lose moistening,and there will be dryness,desquamation,pruritus,and no luster of hands and nails.Therefore,in the general treatment of psoriasis,Qi and Yin should be taken care of.Anemarrhena asphodeloides,Bge.which the cold liquid is thick and smooth,can nourish yin and clear heat,and enrich water to suppress fire.It is recorded in many ancient books that Anemarrhena asphodeloides is often combined with heat clearing and blood cooling drugs to treat skin diseases.In the treatment of psoriasis,there are also classic famous prescriptions such as Baihu Decoction and Xiaofeng powder.On the basis of previous achievements,our team selected Anemarrhena asphodeloides as the intervention drug.We used the method of combining network pharmacology and pharmacological experiments to explore and discuss the pharmacological mechanism of Anemarrhena asphodeloides in the treatment of psoriasis.Objective1.The potential target data set of Anemarrhena asphodeloides in the treatment of psoriasis was constructed by network pharmacology research method,and the biological function and pathway enrichment were analyzed to predict the pharmacological mechanism of Anemarrhena asphodeloides in the treatment of psoriasis.2.Objective To observe the therapeutic effect of Anemarrhena asphodeloides on imiquimod induced psoriasis like model in BALB/c mice.3.To verify part of the pharmacodynamic mechanism predicted by network pharmacology,whether the pharmacodynamic effect is related to apoptosis,and whether the effect of apoptosis plays a role through PI3K/Akt signaling pathway.Methods1.In this study,we first used network pharmacology to integrate a variety of online databases,predict the active ingredients and target information of Anemarrhena asphodeloides,and obtain the relevant targets of psoriasis.The protein interaction(PPI)network model of target data set was constructed by using String database,KEGG and GO were analyzed by using David online database,and the related pathways and biological processes of intersection targets were enriched and analyzed.The interaction network of components,targets and pathways of Anemarrhena asphodeloides was constructed by using Cytoscape software to predict the pharmacological mechanism of Anemarrhena asphodeloides in the treatment of psoriasis.2.Objective to study the effect of Anemarrhena asphodeloides on body weight,spleen index and PASI score in the psoriasis model of BALB/c mice induced by imiquimod.The effect of Anemarrhena asphodeloides on the degree of keratosis and inflammatory cell infiltration of psoriasis model was observed by HE staining,so as to determine whether the decoction of Anemarrhena asphodeloides has the pharmacodynamic effect on imiquimod induced psoriasis model in BALB/c mice.3.The effects of Anemarrhena asphodeloides on apoptosis were studied by TUNEL fluorescence staining and the effects of apoptosis related proteins Bcl-2 and Bax,PI3K/Akt signaling pathway related proteins were detected by Western blot,to verify the partial mechanism of Anemarrhena asphodeloides on imiquimod induced psoriasis in BALB/c mice,whether the effect is related to apoptosis,and whether the effect of apoptosis plays a role through PI3K/Akt signaling pathway.Resultsl.The network pharmacology research obtained 62 intersecting targets and 122 pairs of target interactions.The main biological processes of Anemarrhena asphodeloides in the treatment of psoriasis include apoptotic process,inflammatory response,cell promotion,response to tumor necroautophagy,etc.,the functions of related targets in the treatment of psoriasis are mainly enriched in protein serine/threonine kinase activity,ubiquitin protein ligase binding,protein kinase activity,NADP binding,etc,The main pathways involved are PI3K/Akt signaling pathway,apoptosis pathway,TNF signaling pathway and MAPK pathway.2.In terms of body weight,spleen index and PASI score,there were significant differences between the blank group and the model group(P<0.05).Compared with the model group,there was a trend of healing in each treatment group,in which the effect of Anemarrhena asphodeloides medium dose group was the most obvious.HE staining results showed that compared with the blank group,the model group had obvious inflammatory cell infiltration,and the epidermal dynamics of this group was disordered.But compared with the model group,most of the treatment groups were improved,and the effect of the middle dose Anemarrhena asphodeloides group was the most obvious.3.Statistical TUNEL apoptosis staining showed that there was a significant difference in the percentage of apoptosis between the blank group and the model group(P<0.05).Compared with the model group,the percentage of apoptosis in every treatment group increased,and the difference was statistically significant(P<0.05).The effect of Anemarrhena asphodeloides medium dose group was the most obvious.Compared with the model group,each treatment group could down-regulate the expression of anti-apoptotic protein BCL2,up-regulate the expression of pro-apoptotic protein BAX,and the expression of PI3K protein and AKT protein in the signaling pathway decreased,and the middle-dose group of Anemarrhena asphodeloides was the most obvious,and the difference was statistically significant(P<0.05).Conclusions1.Anemarrhena asphodeloides treatment of psoriasis may be related to a variety of biological processes,cell functions and signal pathways.2.Anemarrhena asphodeloides has therapeutic effect on psoriasis model mice induced by imiquimod,and the middle dose of Anemarrhena asphodeloides has significant therapeutic effect.3.Anemarrhena asphodeloides has a therapeutic effect on imiquimod induced psoriasis model mice,which may be related to apoptosis,which may be partly related to the inhibition of PI3K/Akt signaling pathway.
