Zhimu is the dry rhizome of Anemarrhena asphodeloides Bge.belongs to Liliaceae.It is an important clinical medicinal material embodied in Chinese Pharmacopoeia.Zhimu from Yi County Hebei Province is a traditional authentic material drug called Xiling Zhimu.With the development of health care and excessive using,the condition of wildness Zhimu is on the crisis.Though the quality of cultivate Zhimu is lowness and unsteadied,comparing to the wildness one,the fact is that cultivate Zhimu is becoming to substitute in the market.For the quality and resource crisis of herb medicine,traditional authentic material drug is the best choice.Traditional authentic material drug is the best herb medicine well known to us.Unique inner quality is the basic guarantee for the good clinic curative effect.So,it's important for us to illuminate the inner quality content.Also,it's the real way to make our Chinese traditional herb medicine cause carrying forward and unfading for ever. In the legioned factor that influence the quality of cultivate Zhimu,we selected the dynamic change state of cultivate Zhimu as subject.We detected and analysed the component content of that,and definituded the changing trend rule.Also,we choice Polysaccharide in Zhimu as the new study orientation for quality control.All the results is as follow:(1),Collecting four-year aged Zhimu sample in the fixed testing zone from March to November in the year of 2008,with sarsasapogenin,Anemasaponin AⅢ,neomangiferin,mangiferin in HPLC as target,we depict the dynamic change trend of cultivate Zhimu.HPLC-ELSD method of content determination of sarsasapongenin and Anemasaponin AⅢwith external standard was set up. Precision,stability and recurrence are all up to the mustard.HPLC-UV method of content determination of mangiferin and neomangiferin was set up.The calibrate equation of neomangiferin is:y = 2841.4x - 24.044,r=0.9999(linearity: 0.1047ug~1.2567ug);And that of mangiferin is:y = 3745.9x - 156.08,r= 0.999 (linearity:0.107ug~0.642ug).Precision,stability and recurrence are all up to the mustard too.In study of the dynamic change state,we found:the content of sarsasapogenin reach to tiptop(13.89 mg/g) at the April,fluctuated little until to September,then descended rapidly to the bottom(7.02 mg/g) at the November. To Anemasaponin AⅢ,the content reach to tiptop(13.47mg/g) at the June and descended to the bottom(3.85mg/g) at the August.The content of neomangiferin and mangiferin fluctuated much all the year,what's more,the trend of both dynamic change state converse.(2),We detected the content of total Polysaccharide in Xiling Zhimu:14.33%. After the separating and purification via the HiTrap DEAE FF,Sephacryl S-300, we got four Polysaccharide sample:A-1,A—2,B-1,B—2,from the total Polysaccharide.In the hallmark experiment of high performance liquid gel penetration chromatography,agarose gel electrophoresis and capillary electrophoresis,we got the results that sample A-1,B-1,B—2 are all homogeneous pure polysaccharide.In the molecular weight distribution experiment with high performance liquid gel penetration chromatography,we got the retention time of A-1,B-1,B—2:16.793min,19.92 min,21.814 min, according which,we Calculated the molecular weight are201000,41100,10500 respectively.The distinctness of this article have the following aspects:(1),To reveal the variation trend of cultivate Zhimu920 collected in different periods by multiple indicator analysis.(2),The method of hallmark experiment with high performance liquid gel penetration chromatography,agarose gel electrophoresis and capillary electrophoresis was applied(3),The method of molecular weight distribution experiment with high performance liquid gel penetration chromatography was set up...
