Objective: To establish the quality standard of Anemarrhena asphodeloides Decoction pieces,so as to effectively control the quality of Anemarrhena asphodeloides and provide theoretical basis for clinical application of Anemarrhena asphodeloides;to study and compare polysaccharides,flavonoids and saponins before and after processing of Anemarrhena asphodeloides,to explore the changes of chemical composition of Anemarrhena asphodeloides after wine processing,and to analyze the processing principle;to study the inhibition of Anemarrhena asphodeloides Dec Therapeutic effect of urinary diseases.Methods: The content changes of polysaccharides in Anemarrhena asphodeloides before and after processing were determined by ultraviolet spectrophotometry;a method for determination of flavonoids and saponins in Anemarrhena asphodeloides before and after processing was established by high performance liquid chromatography;the inhibitory effects of decoction,polysaccharides,flavonoids and saponins on alpha-glucosidase before and after processing were determined by PNPG method.Results: The quality standard of Anemarrhena asphodeloides was established.The polysaccharide content of Anemarrhena asphodeloides increased by 1.82%,mangiferin content increased by 0.25%,isomangiferin content increased by 0.11%,saponin BII content decreased by 1.02%,saponin A III content increased by 1.09%,and the water decoction,polysaccharide,total flavonoids and total saponins crude extracts of Anemarrhena asphodeloides processed by alcohol inhibited the activity of alpha-glucosidase.The effect of crude extracts of total flavonoids is more significant than that of raw Anemarrhena asphodeloides.Conclusion: A scientific quality standard of Anemarrhena asphodeloides was established in this experiment.The polysaccharides,mangiferin and saponinAIII of Anemarrhena asphodeloides increased after processing.Alpha-glucosidase inhibitors can inhibit the hydrolysis of glycoside bonds,thus effectively reduce postprandial blood glucose levels,and have a good effect on type II diabetes mellitus.The significant enhancement ofhypoglycemic effect of Anemarrhena asphodeloides may be related to the incremental components of Anemarrhena asphodeloides.This conclusion provides scientific basis and processing mechanism for rational clinical application in the future...
