| Objective: 1.To condense Professor Jiang Yilan's academic thinking on the treatment of colorectal cancer.2.To explore the mechanism of Jianpi Xiaoai Formulas in the treatment of colorectal cancer by network pharmacology and molecule docking.Methods: 1.To explore the origins of academic thought,understand the content of academic thought and collect applied cases through clinical consultations with teachers,study of classics and interviews.2.TCMSP,HERB,TCMDatabase@Taiwan,and TCMID databases were used to obtain the active ingredients of Jianpi Xiaoai Formulas,the active ingredient targets of Jianpi Xiaoai Formulas were obtained from Pharmmapper and Swisstarget Prediction websites respe ctively,and the key genes of Jianpi Xiaoai Formulas against colorectal cancer were obtained using Disgenet,Gene Cards and The key genes for the effect of Jianpi Xiaoai Formulas on colorectal cancer were obtained using Fun Rich3.1.3 software,the Jianpi Xiaoai Formulas-Drug-Active IngredientKey Gene Network was constructed using Cytoscape3.7.2 software,the protein intera ction network was constructed using String database,and the protein interaction netw ork was constructed using The Network Analyzer tool of Cytoscape 3.7.2 software wa s used for network analysis,the metascape website was used for GO analysis of the ke y gene network,and the Cluster Profiler package in R language was used for KEGG p athway enrichment analysis.i GEMDOCK 2.1 software for molecular docking validation,the heatmap package in R and Discovery Studio 2016 Client software for visual presentation of molecular docking results.Results: 1.Professor Jiang Yilan 's academic thinking on the treatment of colorectal cancer includes a discussion of the etiology and pathogenesis of colorectal cancer based on the principle of "stasis and toxicity deficiency",the general pathological phenomenon of "body cold and tumour heat" in colorectal cancer patients,the classification of colorectal cancer into early,middle and late stages,and the focus on the strengthening of the spleen and stomach throughout the whole process of colorectal cancer.In the whole process of colorectal cancer,we pay attention to the protection of the spleen and stomach,and based on this,we have created a formula for colorectal cancer: the formula for strengthening the spleen and eliminating cancer.The treatment of colorectal cancer is a combination of Chinese and Western medicine,with multiple methods of internal and external treatment being used together.We attach importance to the psychological treatment of colorectal cancer patients and regard "practising benevolence with a benevolent heart" as the ultimate care for patients with malignant tumours.2.A total of 50 active ingredients of the formula for strengthening the spleen and elimin ating cancer(ginseng,coix seed,yujin,shigella,vine pear root and snake tongue herb)were obtained after screening,of which 22 were ginseng,9 were coix seed,15 were y ujin,6 were vine pear root and 7 were snake tongue herb,while no eligible active ingr edients of shigella passed the screening.There were 1008 active ingredient target gene s,including 747 for ginseng,156 for coix seed,552 for yujin,171 for vine root and 341 for snake tongue.There were 3168 colorectal cancer disease genes,and 515 key gen es were identified in the effect of Jianshu and Cancer Relief Formula on colorectal ca ncer,and the top ten core genes were: PIK3 CA,PIK3R1,MAPK1,TP53,EP300,SR C,MAPK3,AKT1,APP and STAT3.GO analysis showed that the key genes were associated with malignant pathw ays,positive regulation of metastase activity,growth factor receptors and STAT3.regulation,growth factor receptor and second messenger signaling,trauma response,gastri n signaling pathway,interleukin signaling pathway,positive cell migration,positive c ell death regulation,toxicant response,VEGFAVEGFR2 signaling pathway,hepatitis B,nitrogenation response of cells,lipid metabo lism,apoptotic signaling pathway and other biological processes.KEGG pathway enri chment analysis showed that key genes were associated with PI3 KAkt signaling pathway,small molecule ribonucleic acid in malignancy,proteoglycan r esponse in malignancy,human cytomegalovirus infection,human lymphotropic Tcell virus infection,adhesive spots,oncogenic viruses,Kaposi's sarcoma herpes virus infection,hepatitis B,Fox O signaling pathway,cell cycle,gastric cancer,AGERAGE signaling pathway,prostate cancer,endocrine antagonism,HIF-1 signalling pa thway,EGFR tyrosine kinase inhibitor,pancreatic cancer,small cell lung cancer,chro nic myeloid leukaemia and other signaling pathway are associated.Conclusion: 1.Professor Jiang Yilan 's academic thinking on the treatment of colorectal cancer includes: inheriting Pan,Leading experts in TCM oncology,following "stasis and toxicity deficiency";summarising the disease state,body cold and tumour heat;treating the disease in stages,emphasising the protection of the spleen and stomach;creating new formulas to strengthen the spleen and eliminate cancer;combining Chinese and Western medicine,sharing multiple methods;treating the body and mind together,and being kind-hearted.2.The main active ingredients of Jianpi Xiaoai Formulas may be involved in the positive regulation of transferase activity,signaling of growth factor receptors and second messengers,small molecules in malignant tumo rs by regulating target genes such as PIK3 CA,PIK3R1,MAPK1 and TP53,and regul ating signaling pathways such as PI3 KAkt signaling pathway,malignant tumor pathway,gastrin signaling pathway,interleuk in signaling pathway RNA regulation,proteoglycan response in malignant tumors and other biological processes that play a role in the treatment of CRC.3.This study predicts the active ingredients and mechanism of action of Jianpi Xiaoai Formulas in the treatment of CRC,which provides ideas and references for further in vivo and ex vivo experimental validation and indepth research,and creates a basis for the secondary development and indepth utilization of Jianpi Xiaoai Formulas. |
