Objective:To explore the effect of exosomes derived from human colon cancer HCT116 cells on tumor formation and metastasis in nude mice,and the intervention and mechanism of Jianpi Xiaoai Decoction.Methods:The 48 nude mice were divided into 4 groups according to the random number table method,group A(blank group),group B(model group),group C(exosomes group),and group D(Chinese medicine+exosomes group).Groups C and D were injected with HCT116 exosomes via the tail vein of nude mice,35ug/mouse,every other day,for a total of 14days.At the same time,group D was given Jianpi Xiaoai Decoction solution by intragastric administration at a concentration of 1.365g/ml,0.2ml/mouse,once a day for 14 days.Groups A,B,and C were given an equal volume of normal saline by gavage.The frequency and time of gavage were the same as those in group D.24 hours after the last tail vein injection of exosomes,4nude mice from each group were randomly selected and sacrificed,and liver and lung tissues were taken and fixed with 4%formaldehyde solution for storage.On the 15th day,the prepared HCT116 cell suspension was taken and injected into the tail vein of nude mice in groups B,C,and D at a concentration of 2×10~7/m L and 0.1 m L/mouse.The experiment was terminated on the 30th day,and liver and lung tissues of nude mice were taken,fixed and stored with 4%formaldehyde solution.During the experiment,the mentality,activity,and diet of the nude mice were observed every other day,and the body mass of the nude mice was measured.The liver and lung tissues fixed with 4%paraformaldehyde solution were pathologically sectioned,HE stained,and immunohistochemical to detect TNF-a,IL-1?,and IL-6.Results:1.After continuous injection of exosomes,compared with the blank group,the nude mice in the exosomal group are less energetic,have less activity and diet,and have a slightly reduced body weight(P<0.05);the body weight of the Chinese medicine+exosomes group is slightly lower Increase(P<0.05).Compared with the exosomes group,the activity and diet of the Chinese medicine+exosomes group increased;body mass increased(P<0.05).After injection of HCT116 cells,compared with the model group,the exosomes group was significantly thinner,poorer in spirit,activity and diet were significantly reduced,and body weight was significantly reduced(P<0.05);the body weight of the Chinese medicine+exosomes group increased(P<0.05).Compared with the exosomes group,the activity and diet of the traditional Chinese medicine+exosomes group increased significantly;the body mass increased significantly(P<0.05).2.After HCT116 cells were injected,the liver tissues of the blank group were normal,and the liver tissues of the model group,exosomes group,and traditional Chinese medicine+exosomes group showed obvious metastatic nodules.Compared with the model group,liver metastasis in the exosomes group was significantly increased(P<0.05),and liver metastasis in the traditional Chinese medicine+exosomes group was decreased(P>0.05).Compared with the exosomes group,the metastasis rate of the traditional Chinese medicine+exosomes group was significantly lower(P<0.01).The lung tissues of the blank group were normal,and tumor thrombi were seen in the lung tissues of the model group,exosomes group,and Chinese medicine+exosomes group.The lung metastasis rates in each group were38%in the model group,42%in the exosomes group,and 38%in the traditional Chinese medicine+exosomes group.3.After injection of exosomes,compared with the blank group,the HE staining of the exosomes group showed increased liver inflammation,and the expression of TNF-a,IL-6 and IL-1?in immunohistochemistry increased(P<0.05);Compared with the exosomes group,the inflammatory reaction in the traditional Chinese medicine+exosomes group was reduced,and the expression of TNF-a,IL-6,and IL-1?decreased(P<0.05).After injection of HCT116 cells,compared with the model group,the HE staining of the exosomal group showed a significant increase in liver inflammatory reaction,and the expression of TNF-a,IL-6 and IL-1?in immunohistochemistry was significantly increased(P<0.05);Compared with the exosomes group,the HE staining liver inflammatory reaction in the traditional Chinese medicine+exosomes group was significantly reduced,and the expression of TNF-a,IL-6 and IL-1?in immunohistochemistry was significantly reduced(P<0.05).Conclusion:1.Exosomes derived from human colon cancer HCT116 cells can induce inflammation in the liver tissues of nude mice,regulate the expression of TNF-a,IL-1?,and IL-6,form a pre-metastasis microenvironment,and promote liver tumor formation and metastasis in nude mice..2.Jianpi Xiaoai Decoction can improve the quality of life of nude mice,inhibit the role of exosomes in the inflammatory microenvironment,reduce the expression of TNF-a,IL-1?,and IL-6,thereby inhibiting tumor formation and metastasis in nude mice.
