Effects Of Yangyin Qushihuayu Recipe On Renal Pathology And Apoptosis In Rats With Hyperuricemia Nephropathy

Objective: To study the effects of Yangyin Qushi Huayu Recipe on uric acid(UA),creatinine(Cr),urea nitrogen(BUN),kidney pathology and cell apoptosis in rats with hyperuricemic nephropathy(HN),and to explore the protective effect of Yangyin Qushihuayu Recipe on kidney damage with high uric acid.Methods: Eleven of 54 male SD rats were randomly selected as the blank group,and the remaining rats were administered with potassium oxazinate combined with adenine gavage to establish an HN rat model.After continuous modeling for2 weeks,the HN rat models were randomly divided into model group,high,medium,and low-dose Chinese medicine group,and allopurinol group,and corresponding drug intervention was given for three weeks.After intragastric administration,blood was collected from the abdominal aorta to determine the renal function(UA,Cr,BUN)levels of rats in each group,and analyze the differences between the indexes of each group.At the same time,HE staining was used to observe the pathological changes of the kidneys in each group of rats,and TUNEL staining was used to observe the changes of cell apoptosis in the renal tissues of rats in each group,and calculate the apoptosis index.Results: 1.Observation results of renal function of rats in each group: Compared with the blank group,the UA,Cr and BUN indexes of the model group were significantly higher(P < 0.01);after drug intervention,the UA levels of rats in the intervention group were significantly lower than those in the model group(P< 0.01).Among them,the decrease of UA index in the allopurinol group was better than that in the high,medium and low dose groups of Chinese medicine(P< 0.01),the high and medium dose group of Chinese medicine is better than the low dose group(P<0.05).The Cr levels of the high and medium dose groups of Chinese medicine and the allopurinol group were significantly lower than those of the model group(P<0.05).The BUN levels of the Chinese medicine middle and low-dose groups and the allopurinol group were significantly lower than that of the model group(P<0.05).2.The appearance of rat kidneys in each group and the results of HE staining light microscope observation: The appearance and renal tissue of the model group were more obvious than the blank group.The kidney tissue of the model group showed a large number of renal tubular atrophy,luminal expansion,and internal renal tubules.A large number of urate crystal deposits can be seen,and connective tissue hyperplasia can be seen in the interstitium,accompanied by more lymphocyte infiltration.After the intervention of corresponding drugs,the kidney appearance and pathological damage in the high and medium dose groups of Chinese medicine and the allopurinol group were all reduced to varying degrees,but the improvement in the low dose group was not obvious.3.TUNEL staining light microscope observation results of rats in each group: The apoptosis of kidney tissue in the model group was significantly higher than that in the blank group(P<0.01).Both the high,medium and low dose groups of Chinese medicine and the allopurinol group can significantly improve renal cell apoptosis(P<0.01).Conclusion: Yangyin Qushihuayu Recipe has a certain regulatory effect on the levels of UA,Cr and BUN indicators in HN rats.It can effectively improve the renal function of HN rats,reduce kidney pathological damage,improve renal tissue cell apoptosis,and have a beneficial effect on the kidneys.It has a certain protective effect,and the middle-dose group has relatively better curative effect.
