| Objective: To compare and study the effect and mechanism of Liuwei Dihuang Decoction of different processed dogwoods on postmenopausal osteoporosis,and explore which dogwood products are compatible with Liuwei Dihuang Decoction to have better anti-osteoporosis effect,and try to clarify its mechanism and provide experimental basis for clinical selection of different processed cornus cornus officinalis and Liuwei Dihuang Decoction to prevent PMOP.Methods: Seventy clean-grade female 6-month-old SD rats were divided into blank group,model group and cornus raw products,cornus wine products,cornus steamed products,and cornus salt products respectively which put in Liuwei Dihuang Decoction Wine Cornus Group,Liuwei Dihuang Decoction Salted Cornus Group,Liuwei Dihuang Decoction Steamed Cornus Group,Alendronate Sodium Control(Positive Drug Group),10 in each group.Except for the blank group,the other groups all adopted the method of removing the bilateral ovaries of female SD rats to establish postmenopausal osteoporosis models.Beginning on the 5th day after modeling,each group was given the corresponding dose by continuous intragastric administration for 90 days,once a day.After 90 days,the rat femur bone mineral density(BMD)value,serum estradiol(Estradiol)level,blood calcium and blood phosphorus content were detected by dual-energy X-ray bone densitometer,ELISA method and radioimmunoassay;alkaline phosphatase(ALP),tartrate-resistant acid phosphatase(TRAP),serum osteocalcin(BGP)levels,HE staining method to observe the pathological tissue sections of the femoral head,RT-q PCR method to detect the expression of rat femur OPG,RANK,RANKL m RNA.Results: Compared with the model group,Liuwei Dihuang Decoction in the raw cornel group,the wine cornel group,the salt cornel group,and the steamed cornel group can significantly increase the BMD value and blood phosphorus content of postmenopausal osteoporosis model rats,and repair the fractured femoral head.Trabecular bone,reduce fat vacuoles,reduce the level of ALP(P<0.05),increase the serum estradiol level of model rats(P<0.05),inhibit the weight gain of model rats(P<0.05),and increase model size The expression of OPG m RNA in mouse femur(P<0.05),and the decrease of RANKL m RNA expression in model rats(P<0.05).In addition,the Liuwei Dihuang Decoction in the raw cornel group,the wine cornel group,and the steamed cornel group can reduce the TRAP level of postmenopausal osteoporosis model rats(P<0.05);the Liuwei Dihuang Tang in the wine cornel group can also increase the blood calcium content(P <0.05),Liuwei Dihuang Decoction in the Jiuyu and Salt Zhuyu groups can reduce BGP levels(P<0.05).Conclusion: Different processed Cornus officinalis can improve postmenopausal osteoporosis when combined with Liuwei Dihuang Decoction.The mechanism may be related to its ability to increase serum estrogen levels and regulate bone OPG/RANK/RANKL system,thereby intervening bone metabolism and affecting bone formation.It is related to bone resorption and improving postmenopausal osteoporosis.Among them,the effect of improving bone mineral density is better in the wine cornel group,and the processed product group is better than the raw cornel group;the wine cornel group,the salt cornel group,and the steamed cornel group Liuwei Dihuang Decoction has no significant effect on anti-postmenopausal osteoporosis According to the difference,the clinic can choose appropriate processed products to be combined with Liuwei Dihuang Decoction to prevent and treat postmenopausal osteoporosis. |
