| Iron Deficiency Anemia(IDA)is caused by various reasons which lead to the lack of iron element stored in body.Iron supplement agents were usually selected to treat IDA,however,the existing iron supplements,there are some side effects,besides their effects of improve anemia are limited.,it is needed to seek safe,efficient and low side effects substance for anemia.Our previous studies have found that Auricularia auricula-judae melanin has significant effect on treating the IDA,however,the specific effectiveness of the composition is not clear,need further study to verify.In this paper,Auricularia auricula-judae melanin as raw material by dextran gel Sephadex G-100chromatography separated three components,namely,Auricularia auricula-judae melanin F1,Auricularia auricula-judae Melanin F2,and Auricularia auricula-judae melanin F3.Iron deficiency anemia in mice fed by blood and hematopoietic factors related indicators to filter main effect components and physicochemical properties of these three components were studied,and the structure of the efficacy of the components was obtained,the results are as follows:(1)IDA+F1 group had a significantly higher number of Hb and RBC compared with IDA and IDA+F2,3 group;The serum SCF concentration,serum EPO,serum GATA-1,serum iron of IDA+F1 group were significantly higher than that of IDA and IDA+F2,3group(p<0.05),among the detected blood indexes the Auricularia auricula-judae melanin F1 groups were superior to Auricularia auricula-judae melanin F2,3 groups,aria auricula-judae melanin F2,3 groups were statistically significant differences(p<0.05).Description Auricularia auricula-judae melanin F1 is to improve the effectiveness of iron deficiency anemia main ingredient.(2)Light will reduce the stability of the components of the Auricularia auricula-judae melanin;cold conducive to the preservation of the components of Auricularia auricula-judae melanin,prolonged heating will reduce the preservation rate of each component of Auricularia auricula-judae melanin;p H value significantly affect the stability of the components of Auricularia auricula-judae melanin,alkaline conditions favor the dissolution of the components of Auricularia auricula-judae melanin;K+,Ca2+and Mg2+can significantly reduce the absorbance value of each component of Auricularia auricula-judae melanin,Cu2+and Fe3+increase the absorbance values of each component of Auricularia auricula-judae melanin;Auricularia auricula-judae melanin components are easily oxidized,but reducing agent had no significant effect on its stability;sodium benzoate,potassium sorbate,sodium glutamate,and butylated hydroxy anisole of the components of Auricularia auricula-judae melanin stability have little effect.(3)Auricularia auricula-judae melanin F1 with pyrrole ring and indole ring skeleton,the skeleton is connected to aliphatic carbon chain and a carbonyl group,after the formation of a large number of irregular polymerization of monomers of these structures,the monomers to form the laminated polymeric composition than the surface smooth irregular Auricularia auricula-judae melanin F1.Strong absorption peak at 207nm.Based on the content of the Auricularia auricula-judae melanin F1 S element calculation,beumelanin and phaeomelanins ratio of 10.36:1 in Auricularia auricula-judae melanin F1.In this paper,through the establishment of iron-deficiency mouse model to determine the Auricularia auricula-judae melanin to improve the effectiveness of iron deficiency anemia in mice group was divided into Auricularia auricula-judae melanin F1,and to determine the basic skeleton structure whose chemical groups,experiments show that the physicochemical properties of Auricularia auricula-judae melanin F1melanoma stability is better than the other two components,for the food and drug safety research and development of new follow-up treatment of iron deficiency anemia foundation. |
