Radix Angelica Dahurica is the dried root of Angelica dahurica(Fisch.ex Hoffm.)Benth.et Hook.f.or Angelica dahurica(Fisch.ex Hoffm.)Benth.et Hook.f.var.formosana(Boiss.)Shan et Yuan(Umbelliferae).Being recorded in Chinese Pharmacopeia 2015 edition,it is classified as a warm herb,which tastes pungent and has effects of dispersing superficial and wind syndromes,drying damp evils,etc.It is commonly-used in treatments of encephalalgia,odontalgia,rhinorrhoea,leukorrhagia,furunculosis and so on.On the basis of previous study of our team,a rapid HPLC-MS-oriented separation combined with various of column chromatography and preparative HPLC was conducted on Radix Angelica Dahurica.Eleven new dimeric furanocoumarins were obtained from the partitions of petroleum ether and EtOAc,and named dahuribiethrin A-J(1-10)and dahurinol A(11).In order to clarify the effective substances of Radix Angelica Dahurica more thoroughly,a systematic study of its chemical constituents in the polar fraction was performed in this project.Up to now,seven pyrrolic alkaloids including five new compounds dahuriine A-E(12-16),and two known compounds butyl(2-formyl-5-butoxymethyl-1Hpyrrole-1)butanoat(17)and hemerocallisamine II(18)were isolated.One phenanthrene dehydroeffusol(19)was also isolated from H2O and n-butanol soluble fraction.In addition,13 known coumarins including imperatorin(20),isoimperatorin(21),alloimperatorin(22),xanthotoxin(23),xanthotoxol(24),bergapten(25),isopimpinellin(26),phellopterin(27),isooxypeucedanin(28),isodemethylfuropinarine(29),demethylsuberosin(30),nodakenin(31),marmesinin(32).Two sterols,daucosterol(33)and ?-sitosterol(34),and p-hydroxy benzaldehyde(35)were isolated and indentified as well.On the biological activities of dimeric furanocoumarins,compounds 2,3,4,5,9,10 exhibited inhibition of nitric oxide production in the lipopolysaccharide(LPS)-stimulated RAW 264.7 macrophage cells with IC50 values of 9.6±0.3,9.8±0.2,8.8±0.4,9.2±0.2,8.7±0.6 and 27.3 ± 0.9(?M),respectively.On the basis of chemical research,we established the HPLC fingerprint of Radix Angelica Dahurica by using 13 batches medicinal materials from different areas.And two common peaks,imperatorin and isoimperatorin,were identified by reference standards.The similarity scores between 14 batchs including one reference crude herb ranged from 0.963-0.993.Besides,byakangelicol and oxypeucedanin were isolated and identified in the utility of HPLC-MS-oriented separation technique,which showed great discrepancy in the contents between 14 batchs.And the ternary oxygen ring may well be transformed in the processing and storage of Angelica dahurica,which leads to the formation of byakangelicin and oxypeucedanin hydrate.The results provide a basis for evaluating the quality of Radix Angelica Dahurica crude drug and a useful reference in its industrialization.
