Objective To investigate the changes of intestinal mucosal immune function in experimental strangulated intestinal obstrction rats and the regulatory effect of Thymosin alpha 1.Methods Forty-eight male S-D rats were randomly divided into A group(sham-operation group),B group(strangulated intestinal obstruction group)and C group(Thymosin alpha 1 group).Animals of A group and B group were given subcutaneous injection of 0.1ml physiological saline and rats in C group were given subcutaneous injection of Thymosin alpha 1 0.16mg per 24 hours.Four days later,rats in A group underwent a sham-operation and animals in the other groups were submitted to strangulated intestinal obstruction.The animals were given drugs after the operation sequentially.Rats were sacrificed at 72h after the strangulated intestinal obstruction models were made.Endotoxin concentration in abdominal aorta and secretory immunoglobulin A in terminal ileum mucosa were determined by Enzyme-linked Immunosorbent Assay.CD3+,CD4+,CD8+T lymphocytes in intestinal mucosal lamina propria were examined by immunohistochemistry.Results(1)The mortality of the rats of A group,B group and C group were 0%,55%and45%.There was no significant difference between them,tested by Kruskal-Wallis test,P>0.05.(2)Compared with A group,B group had a lower CD3+,CD4+,CD8+T lymphocyet percentage in the end of intestimal mucosal lamina propria and lower s Ig A concentrations in caecum intestinal mucosa and higher level of plasma endotoxin concentration in the abdominal aorta(F=12.63-30.00,t AB=4.54-7.78,P<0.05).(3)Compared with B group,C group had a lower level of plasma endotoxin concentration in the abdominal aorta(F=14.84,t BC=3.41,P<0.05)and a higher CD3+,CD4+,CD8+T lymphocyet percentage in the end of intestimal mucosal lamina propria(F=12.63-30.00,t BC=4.08-4.21,P<0.05).The C group had a higher level of s Ig A concentrations in caecum intestinal mucosa,and the difference between B group and C group was not significantly(F=15.19,t BC=1.90,P>0.05).Conclusion The intestinal immune function of strangulated intestinal obstruction in rats decreased,increased bacterial translocation.of thymosin alpha 1 can improve the intestinal immune function and reduce endotoxin translocation in the strangulated intestinal obstruction rats.
