N.baileyi,also known as the red goral,belongs to the Artiodactyla Bovidae Caprinae Naemorhedus.Protection of animals at the national level.It is classified as vulnerable by IUCN and belongs to CITES Appendix 1 species.The wild N.baileyi is distributed in a narrow area and the population is sparse.However,the captive N.baileyi currently only exists in the Shanghai Zoo.It is extremely difficult to carry out systematic research on the N.baileyi.There are 12 N.baileyi individuals in Shanghai Zoo,but the number of suitable breeding individuals is only 3(2 males and 1 female).There is still a serious imbalance in the sex ratio within the structure,and the number of individuals has been declining in recent years.in order to increase the breeding rate of N.baileyi and expand the number of individuals in the more this is necessary to adopt scientific auxiliary measures to increase the matching rate In the past,observing the estrus behavior of the N.baileyi was more subjective,which not only disrupted the original estrus period of the N.baileyi,but may also cause unnecessary fighting casualties in extreme cases.The determination of the content of reproductive hormones in feces can more objectively reflect the individual’s estrus state,which is an ideal way to predict the estrus period.Therefore,we hope to establish a continuous collection of rubella faeces,effective storage and determination of reproductive hormone content in fecal samples,combined with traditional manual observations,objectively and accurately predict the estrus period,ensure the effectiveness of the cage,and reduce the captive N.baileyi.Psychological and physical damage.Under certain field conditions or captive conditions,it may not be possible to process samples in time.Finding a good preservation method is one of the prerequisites for fecal hormone research.Studies on a variety of animal fecal samples have shown that different storage times and storage methods can lead to large differences in fecal hormone levels.Therefore,it is very important to study the time-effect of hormone levels in the stool for the N.baileyi.This experiment verifies the effect of different storage methods on the timeliness of fecal hormones.Collect samples of rubella feces and perform five treatments:(1)No treatment,(2)95% absolute ethanol,(3)Levofloxacin lactate antibiotic,(4)50°C oven drying,(5)-20°C Freezer freezing,all groups are at room temperature except for freezing treatment.And from 4 time gradients(15d,30 d,90d,150d)to test the changes of estradiol,progesterone,and testosterone in the stool of each gradient.The results show that different hormones require different storage methods.Estradiol can be stored in four ways except antibiotics.Among them,ethanol is short-acting storage(<30d),and the best choice is drying or freezing.Progesterone can be used in four other ways except antibiotics,and the best choice is freezing.Testosterone can use the other four methods except antibiotics,among which freezing is short-acting storage(<30d),and the best choice is no treatment.In actual processing,it can be treated flexibly according to experimental conditions and instruments.For the prediction of the estrus period of the N.baileyi,this study collected the fecal samples of two reproductive-age N.baileyi(1 female and 1 male)in Shanghai Zoo in 2018 and 2019.The feces of the female N.baileyi were tested for the content of estradiol and progesterone.Determination of male N.baileyi feces to determine the content of the reproductive hormone testosterone,draw a graph to observe the annual fluctuations of the reproductive hormone,and reasonably estimate the estrus period to assist in judging the timing of cage closure.According to the annual fluctuation of estradiol concentration,it is reasonable to predict the estrus period of female N.baileyi in the middle and late January,late November to mid-December.According to the annual fluctuation of progesterone concentration,the estrus period of female N.baileyi is reasonably predicted to be in early January,late January,early November to mid-December.According to comprehensive judgment,the predicted estrus period of female N.baileyi is the end of January and mid-December.According to the annual fluctuation of testosterone concentration,it is reasonable to predict that the estrus period of male N.baileyi will be from early January to early March,with more intense mid-to-early January,mid-April to mid-May,and mid-October to mid-late November.Under ideal conditions,the two periods of end of January and end of November are both the estrus period of male and female N.baileyi.Theoretically,the successful rate of co-cage is the highest during this period.Due to the large amount of uncertainty in actual conditions,in actual production and life,the combination of expanding the cage time and observing estrus behavior can be used to maximize the breeding rate while ensuring the safety of N.baileyi.
